Abstract
Penelitian studi kadar kuersetin pada “teh” batang daun alga coklat Sargassum cristaefolium telah dilakukan. Metode penelitian yang digunakan adalah metode eksploratif deskriptif dengan tujuan yang utama untuk mengetahui kadar kuersetin dari batang daun alga coklat Sargassum cristaefolium. Perlakuan yang diberikan pada alga coklat Sargassum cristaefolium yaitu kondisi segar, kering (alga yang dikeringkan dengan sinar matahari selama 2x24 jam), “teh” (alga coklat direndam dengan Ca(OH)2 pH 11 selama 6 jam dan dikeringkan dengan vacuum dryer suhu 80oC selama 29 menit), dan “teh seduh” berasal dari sampel “teh” yang diseduh dengan menggunakan air panas. Dalam penelitian alga coklat Sargassum cristaefolium diuji kadar kuersetin dengan larutan pengekstrak metanol. Kadar kuersetin dari ekstrak diukur dengan High Performance Liquid Chromatography (HP-LC), sedangkan aktivitas antioksidan diukur menggunakan 1,1-dyphenil-2-picrylhydrazylDPPH. Hasil penelitian menunjukkan bahwa kadar kuersetin ekstrak alga coklat kering lebih inggi daripada eksrak alga coklat segar, “teh”, dan “teh seduh” dengan kadar kuersetin berturut-turut 0,359 µg/ml, 0,112 µg/ml, sampel segar dan “teh seduh” tidak ditemukan. Hasil penelitian juga menunjukkan bahwa aktivitas antioksidan ekstrak alga kering lebih tinggi daripada ekstrak alga segar, “teh”, dan “teh seduh” dengan nilai IC50 berturut-turut 157,17 ppm, 169,91 ppm, 185,48 ppm, dan 249,62 ppm.
English Abstract
Research about quercetin content in “tea” mixture of brown algae Sargassum cristaefoliumleaves and thallus had been done. The research used descriptive explorative method with the main purpose is to know the quercetin content in “tea” mixture of brown algae Sargassum cristaefolium leaves and thallus from brown algae. Fresh brown algae Sargassum cristaefolium, sundried (brown algae was dried in the sun for 2x24 hours), “tea” (brown alga soaked in a solution of Ca(OH)2 pH 11 for 6 hours and dried using a vacuum dried 80oC for 20 minutes) and “tea” solution from “tea” samples brewed using hot water. The samples were analyzed to know the quercetin content with methanol solvent extracted. Quercetin content from extracts were determined by High Performance Liquid Chromatography(HP-LC), while the antioxidant activity was measured using 1,1-dyphenil-2-picrylhydrazyl (DPPH). The result showed that content quercetin of dried algae extract was higher than fresh algae, “tea”, and “tea solution”. It showed that quercetin content: of dried Sargassum cristaefoliumwas 0,359 µg/ml, while “tea” Sargassum cristaefolium was 0,112 µg/ml but, quercetin content in “fresh” and “solution tea” were undetected. The result also showed that the antioxidatint activity of sundried algae extract was higher then fresh alga, “tea”, and “tea” solution with IC50 of the treatments in 157,17 ppm, 169,91 ppm, 185,48 ppm, dan 249,62 ppm..
