Abstract

Diabetes Mellitus (DM) tipe 1 merupakan penyakit yang disebabkan adanya kelainan sekresi insulin. Defisiensi insulin dapat menyebabkan peningkatan kadar Low Density Lipoprotein (LDL) dan kerusakan pada hepar. Tujuan penelitian ini adalah untuk mengetahui kadar LDL dan kerusakan hepar pada tikus (Rattus norvegicus) model diabetes mellitus tipe 1 hasil induksi streptozotocin (STZ) yang diberi terapi ekstrak ethanol kunyit (Curcuma Longa L). Rancangan penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Penelitian ini menggunakan tikus (Rattus norvegicus) jantan berumur 3 bulan yang dibagi dalam 5 kelompok yaitu: (A) kelompok kontrol negatif (tidak diinduksi STZ), (B) kelompok kontrol positif (diinduksi STZ / tikus DM 1), (C) kelompok tikus DM 1 dengan terapi ekstrak ethanol kunyit dosis 1,2 g/kg BB, (D) kelompok tikus DM 1 dengan terapi ekstrak ethanol kunyit dosis 1,8 g/kg BB, dan (E) kelompok tikus DM 1 dengan terapi ekstrak ethanol kunyit dosis 2,7 g/kg BB. Masing – masing kelompok terdiri atas 4 ekor tikus. Induksi STZ secara intraperitonial dengan dosis 20 mg/kg berat badan selama 5 hari. Terapi dilakukan dengan cara sonde lambung selama 6 minggu. Kadar LDL diukur dengan metode direct dan tingkat keparahan kerusakan hepar diamati melalui gambaran histopatologinya. Hasil penelitian menunjukkan bahwa pemberian ekstrak ethanol kunyit dosis 2,7 g/kg BB pada tikus model diabetes mellitus tipe 1 mampu menurunkan kadar LDL sampai 59,55% dan menurunkan tingkat keparahan perlemakan hepar dengan berkurangnya vakuola lemak. Kesimpulan dari penelitian ini adalah bahwa ekstrak ethanol kunyit mengandung antioksidan yang mampu menurunkan kadar LDL dan menurunkan tingkat keparahan perlemakan hepar.

English Abstract

Diabetes Mellitus (DM) type 1 is a disease caused by abnormalities in insulin secretion. Insulin deficiency can increase level of Low Density Lipoprotein (LDL) and damage the liver. This research was conducted to determine level of LDL and hepatic damage in type 1 diabetes mellitus rat (Rattus norvegicus) model induced by streptozotocin which was treated with turmeric ethanol extract. This experiment was design by Completely Randomized Design. This study used 3-month-old male rats (Rattus norvegicus) which were divided into 5 groups: (A) negative control group (not induced by STZ), (B) positive control group (STZ-induced / DM Type 1 rats), (C) DM Type 1 rats treated with 1,2 g /kg BW dose of turmeric ethanol extract, (D) DM Type 1 rats treated with 1,8 g/kg BW dose of turmeric ethanol extract and (E) DM Type 1 rats treated with 2,7 g/kg BW dose of turmeric ethanol extract. Each group consisted of 4 rats. Induction of DM Type 1 in rats was done by injecting 20 mg/kg BW Multiple Low Dose - Streptozotocin (MLD-STZ) intraperitonially (i.p) in 5 days. Therapy was given using stomach probe for 6 weeks. Low Density Lipoprotein level was measured by the direct method and hepatic damage was examined through histopathology image. The results showed that 2.7 g/kg BW dose of turmeric ethanol extract in rat model of type 1 diabetes mellitus can decrease LDL level up to 59.55% and reduce the severity of fatty liver with reduced fat vacuoles. As a conclusion, turmeric ethanol extract contains antioxidants that can decrease LDL level and reduce the severity of fatty liver.
