Abstract

Diabetes mellitus adalah kelainan sistem endokrin yang mendorong laju metabolisme kompensatorik terutama pada hati, otot rangka, dan jaringan lemak. Pengidap diabetes memiliki kerentanan terhadap berbagai komplikasi yang serius dan progresif. Predisposisi penyakit kardiovaskular pada pengidap diabetes disebabkan adanya gangguan pada produksi dan eliminasi komponen lipid, terutama low density lipoprotein (LDL). Faktor pemicu stres oksidatif seperti meningkatnya reactive oxygen species (ROS) dapat berinteraksi dengan LDL, menghasilkan ox-LDL yang tidak dikenali reseptornya sehingga kadar LDL sirkulasi meningkat. Ipomoea batatas L. adalah tanaman umbi dengan kandungan pigmen antosianin sebagai scavenger radikal bebas. Antosianin juga berperan dalam mencegah resistensi insulin dan menghambat lipolisis. Penelitian eksperimental ini menggunakan Post Test-Only Control Group Design untuk mengetahui pengaruh ekstrak antosianin ubi jalar ungu (Ipomoea batatas L.) terhadap kadar LDL. Penelitian dilakukan terhadap tikus wistar (Rattus norvegicus) jantan (n=20) yang dibagi ke dalam 5 kelompok: 3 kelompok dosis ekstrak antosianin (10 mg/kgBB, 20 mg/kgBB, 80 mg/kgBB) dan 2 kelompok kontrol (normal dan diabetes mellitus). Induksi diabetes mellitus dilakukan dengan pemberian diet tinggi lemak dan injeksi streptozotocin. Perlakuan dilakukan selama 47 hari. Hasil uji One-way ANOVA menunjukkan terdapat perbedaan rata-rata yang bermakna terhadap kadar LDL tikus (p=0,041). Dari uji LSD Post Hoc didapatkan penurunan kadar LDL yang bermakna pada pemberian ekstrak antosianin dosis 20 mg/kgBB (p=0,025). Tidak didapatkan hubungan yang bermakna antara kenaikan dosis ekstrak antosianin dengan penurunan kadar LDL tikus (p=0,170, r=0,361). Dengan demikian dapat disimpulkan bahwa ekstrak antosianin ubi jalar ungu (Ipomoea batatas L.) dapat menurunkan kadar LDL tikus wistar yang mengidap diabetes mellitus dengan pemberian dosis 20 mg/kgBB.

English Abstract

Diabetes mellitus is an endocrine system disorder driving compensatory metabolism, especially in liver, sceletal muscles, and adipose tissue. Patients are susceptible to progressive and serious complications. Predisposition of cardiovascular disease on the diabetics is caused by production and elimination disturbance on the lipid component, especially low density lipoprotein (LDL). Oxidative stress triggers such as the increasing reactive oxygen species (ROS) interacting with LDL, results in unknown ox-LDL to its receptor, which leads to the increasing level of circulating LDL. Ipomoea batatas L. is a functional nutraceutical containing anthocyanin pigment working as free radical scavenger. Anthocyanin also prevents from insulin resistance and inhibits lipolysis. An experimental research using Post Test-Only Control Group Design is carried out to identify the effect of anthocyanin extract of purple sweet potato (Ipomoea batatas L.) on LDL level. The experiment is applied to male wistar rats (Rattus norvegicus) (n=20) divided in 5 groups: 3 groups applied dose of anthocyanin extract (10 mg/kgBW, 20 mg/kgBW, 80 mg/kgBW) and 2 control groups (normal and diabetes mellitus). The induction of diabetes mellitus is done by applying high fat diet and streptozotocin injection. The intervention is completed within 47 days, One-way ANOVA test shows that there is a significant mean difference on the LDL level in rats (p=0.041). LSD Post Hoc test results in significant decrease of LDL level after the application of anthocyanin extract dose of 20 mg/kgBW (p=0.025). There is no significant correlation between the increase of anthocyanin extract dose with the decrease of LDL level in rats (p=0.170, r=0.361). It can be concluded that the anthocyanin extract of purple sweet potato (Ipomoea batatas L.) applied in dose of 20 mg/kgBW, decreases LDL level of wistar rats having diabetes mellitus.
