Abstract
Infeksi merupakan penyebab kematian ibu tertinggi ketiga di Indonesia. Bakteri penyebab infeksi antara lain Staphylococcus aureus. Salah satu penyakit yang disebabkan oleh bakteri Staphylococcus aureus adalah mastitis. Penyakit ini berpengaruh pada kualitas ASI yang diberikan pada bayi. Buah mahkota dewa (Phaleria macrocarpa) merupakan tanaman yang banyak ditemui di Indonesia. Buah ini memiliki zat aktif flavonoid sebagai antimikroba, tetapi masyarakat belum banyak yang memanfaatkannya. Penelitian ini bertujuan untuk mengetahui potensi antimikroba ekstrak flavonoid buah mahkota dewa terhadap bakteri Staphylococcus aureus secara in Vitro. Sampel diperoleh dari isolate Staphylococcus aureus di Laboratorium Mikrobiologi FKUB. Konsentrasi ekstrak penelitian yang digunakan yaitu 3.5%, 3%, 2.5%, 2%, 1.5%, 1%, 0.5%, dan 0% (KK). Metode penelitian menggunakan dilusi tabung dengan tiga kali pengulangan pada setiap konsentrasi. Hasil statistic One-Way ANOVA menunjukkan terdapat perbedaan yang signifikan pada perubahan konsentrasi ekstrak flavonoid buah mahkota dewa terhadap biakan Staphylococcus aureus (p<0.05). Uji korelasi dan regresi menunjukkan adanya hubungan yang erat dan berbanding terbalik serta pengaruh yang signifikan antara konsentrasi ekstrak dengan jumlah koloni bakteri (Korelasi, r= -0.561; p<0.05; dan R square 31.5%). Berdasarkan penelitian ini, dapat disimpulkan bahwa ekstrak flavonoid buah mahkota dewa mempunyai efek antimikroba terhadap bakteri Staphylococcus aureus dengan KHM 1% dan KBM 2%.
English Abstract
Infection is the third highest cause of maternal mortality in Indonesia. The bacteria that cause infections include Staphylococcus aureus. One of the diseases caused by the Staphylococcus aureus is mastitis. These diseases affect the quality of breast milk fed to infants. Mahkota dewa (Phaleria macrocarpa) is a plant that is found in Indonesia. The fruit has flavonoids as an antimicrobial active agent, but not many people use it. This study aims to determine the antimicrobial potency flavonoid extract of mahkota dewa fruit to against Staphylococcus aureus in Vitro. Samples were obtained from Staphylococcus aureus isolates in the Laboratory of Microbiology FKUB. The extract used in this study is 3.5%, 3%, 2.5%, 2%, 1.5%, 1%, 0.5%, and 0% (KK). The research method using a dilution tube with three repetitions at each concentration. Results statistic of One-Way ANOVA Showed a significant difference in the change on concentration of flavonoids extract againts Staphylococcus aureus (p <0.05). Correlation and regression analysis showed a close relationship and inversely, as well as having a significant difference between the concentration of the extract by the number of bacterial colonies (correlation, r = -0561; P> 0.05; R square and 31.5%). Based on this study, it can be concluded that the flavonoid extract of Phaleria macrocarpa has an antimicrobial effect against Staphylococcus aureus with the Minimum Inhibitory Concentration is 1% and Minimum Bactericidal Concentration is 2%.
