Abstract

MDA atau Malondialdehyde adalah marker pertanda adanya proses peroksidasi lipid yang terjadi di jaringan hati yang menyebabkan peningkatan kondisi stres oksidatif, yang akan mengaktivasi hepatic stellate cells (HSC), meningkatkan kadar sel matriks ekstraseluler dalam jaringan hati, dan meningkatkan deposisi kolagen pada jaringan yang rusak, yang berakibat adanya kondisi fibrosis hati. Kurkumin diyakini memiliki efek anti fibrosis dengan mekanismenya yang dapat menekan aktivasi dari HSC dan juga dapat meningkatkan kadar PPAR-γ sehingga akan menekan kadar NF-kB yang berefek pada peningkatan apoptosis dari sel-sel yang telah rusak, sehingga kadar ROS pada jaringan hati akan berkurang. Penelitian ini ingin mengetahui pengaruh lama pemberian kurkumin dengan penurunan kadar MDA jaringan hati pada tikus model fibrosis hati akibat induksi karbon tetraklorida. Sampel dibagi menjadi 8 kelompok yang terdiri dari kontrol negatif, kontrol positif, kelompok perlakuan dengan lama pemberian kurkumin selama 2 minggu (KP2) dan kelompok kontrolnya (KK2), kelompok perlakuan dengan pemberian kurkumin selama 5 minggu (KP5) dan kelompok kontrolnya (KK5), kelompok perlakuan dengan pemberian kurkumin selama 9 minggu (KP9) dan kelompok kontrolnya (KK9). Dari hasil penelitian ini, didapatkan perbedaan yang bermakna antara 8 kelompok perlakuan ini (one way ANOVA p=0,000) pada uji korelasi Pearson, hubungan antara lama pemberian kurkumin dengan penurunan kadar MDA jaringan hati memiliki korelasi yang kuat, signifikan, dan bersifat dua arah.

English Abstract

MDA or Malondialdehyde is a potential marker of lipid peroxidation happened in the liver tissue that causes an increasing level of oxidative stress condition, that will activate the hepatic stellate cells (HSC), an increasing level of extracellular matrix in liver tissue, and increasing level of collagen deposition in the injured tissues, these three processes soon will cause a liver fibrosis. Curcumin is believed to has an antifibrotic effect by suppressing the activation of HSC and increasing the level of PPAR-γ that will suppress the level of NF-kB, this process will affect in the increasing level of the injured cells’s apoptosis and the decreasing level of ROS in liver tissue. This study is aimed to test the effect of the duration of curcumin therapy on the decreased level of malondialdehyde in mice’s liver tissue induced by carbon tetrachloride. Samples are divided into 8 groups that consist of negative control, positive control, the curcumin-given treatment group for 2 weeks and it’s control, the curcumin-given treatment group for 5 weeks and it’s control, and the curcumin-given treatment group for 9 weeks and it’s control. The result of this experiment shows that there is a significant difference between the 8 groups of samples (One Way ANOVA p=0,000) with normal and homogenous data distribution. From the Pearson correlation test, the correlation between the duration of curcumin therapy and the decreasing level of MDA liver tissue has a significant, two-way, and strong correlation.
