Abstract
Latar Belakang : Susu sapi bubuk mengandung berbagai komponen di antaranya kalsium dan vitamin D yang diduga berpengaruh terhadap ketebalan dinding aorta pada DM tipe 2. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian susu sapi bubuk terhadap ketebalan dinding aorta pada tikus wistar model DM tipe 2. Metode : Penelitian ini merupakan penelitian eksperimental dengan desain Post Test Only Control Group. Pemilihan sampel dilakukan menggunakan simple random sampling dengan total sampel 30 ekor tikus wistar jantan yang dibagi menjadi 5 kelompok : K(-) (diet normal), K(+) (diet tinggi lemak + induksi STZ), P1 (diet tinggi lemak + induksi STZ + susu sapi bubuk 0,9 g), P2 (diet tinggi lemak + induksi STZ + susu sapi bubuk 1,8 g), dan P3 (diet tinggi lemak + induksi STZ + susu sapi bubuk 2,7 g). Semua tikus wistar diadaptasi 1 minggu dengan diet normal kemudian dilanjutkan diet tinggi lemak selama 4 minggu. Pada hari ke-94 ketebalan dinding aorta tikus diukur dengan pembesaran 400x. Data diolah dan dianalisis menggunakan SPSS 16. Uji statistik menggunakan uji korelasi Spearman. Hasil : Hasil penelitian menunjukkan adanya hubungan yang signifikan antara pemberian susu sapi bubuk dengan ketebalan dinding aorta yang diukur dari tunika intima sampai tunika media, dengan kekuatan korelasi sedang dan arah korelasi positif (p = 0,008 dan R = 0,519). Kesimpulan : Hasil penelitian ini tidak sesuai dengan hipotesis penelitian, di mana pemberian susu sapi bubuk dengan dosis 0,9; 1,8; dan 2,7 gram/hari selama 8 minggu justru secara signifikan berpengaruh terhadap peningkatan ketebalan dinding aorta pada tikus dengan DM. Diperlukan penelitian lanjutan dengan jumlah sampel yang lebih besar dan diaplikasikan pada manusia.
English Abstract
Background: dairy milk powder contains of various components such as calcium and vitamin D. These components are thought to be able to affect the arterial wall in type 2 diabetes mellitus. The aim of this study was to determine the effect of powdered milk to the aortic wall thickness of Wistar rat model of type 2 diabetes mellitus. Methods: This was an experimental study using Post Test Only Control Group Design. Sampling was carried out by using simple random sampling with 30 rats for total sample which were divided into five groups: K(-) (normal diet without STZ induction), K(+) (high fat diet + STZ induction), P1 (high fat diet + STZ + 0,9 g milk powder), P2 (high fat diet + STZ + 1,8 g milk powder), and P3 (high fat diet + STZ + 2,7 g milk powder). All rats had been adapted to the normal diet for 1 week, then continued with high fat diet for 4 weeks. On the 94th day rats’ aortic walls thickness were measured at 400x magnification. Data were processed and analysed using SPSS 16, the statistical test was using Spearman correlation test. Results: The test showed a significant correlation between dairy milk powder intervention to the aortic wall thickness as measured from tunica intima to tunica media, with positive correlation and the strength of the correlation is moderate as shown by p = 0,008 and R = 0,519. Conclusion: This experimental study showed different result with the hypothesis. The deliverance of dairy milk powder with some doses such as 0,9; 1,8; and 2,7 g/day for 8 weeks affected the increasing of aortic thickness in rats with diabetes mellitus significantly. Further research is needed with larger sample size and its application to humans.
