Abstract
Aterosklerosis adalah penyakit kronis yang ditandai oleh akumulasi lemak dalam dinding arteri yang akhirnya membentuk plak yang dapat menyebabkan penyempitan, pengerasan dan atau penyumbatan lengkap dinding arteri. BAT merupakan jaringan yang kaya akan mitokondria berperan dalam penurunan timbunan lemak adiposa melalui reseptor transkripsi gen PPARs α serta mensekeresi anti inflamasi adiponektin yang dapat menghambat terbentuknya plak aterosklerosis. Pada suatu penelitian untuk membentuk suatu kondisi aterosklerosis diperlukan diet aterogenik modifikasi standar AIN-93M yang merupakan high fat diet modifikasi AIN-93M dengan penambahan asam kolat dan kolesterol. Penelitian ini bertujuan untuk mengetahui perbedaan antara pemberian diet aterogenik modifikasi standar AIN-93M dengan diet normal standar AIN-93M terhadap jumlah Brown Adiposa Tissue tikus Rattus Norvegicus Strain Wistar jantan. Penelitian ini menggunakan true experimental laboratory dengan metode Randomized Control Group Postest Design dengan menggunakan tikus Rattus Novergicus Galur Wistar jantan. Pemilihan sampel dilakukan menggunakan metode RAL (Rancangan Acak Lengkap) dengan jumlah sampel total sebanyak 32 tikus. Uji statistik menggunakan independent T test. Hasil penelitian menunjukan kelompok diet aterogenik modifikasi standar AIN-93M (P1) dengan rata-rata asupan 8,6 ± 2,01 gram, energi 37,1 ± 8,6 kkal, dan jumlah Brown Adiposa Tissue 0,15 ± 0,27 gram sedangkan kelompok diet normal standar AIN-93M (P2) dengan rata-rata asupan 10,6 ± 1,27 gram, energi 35,6 ± 4,26 kkal, dan jumlah Brown Adipose Tissue 0,12±0,35 gram . Peningkatan BAT dalam diet aterogenik (P1) diduga sebagai mekanisme homeostasis tubuh terhadap kondisi hiperkolesterolemia tanpa dsertai obesitas untuk mengurangi akumulasi lemak pada jaringan adiposa dan mencegah terjadinya toksisitas lemak. Berdasarkan penelitian ini dapat disimpulkan ada perbedaan yang signifikan jumlah Brown Adipose Tissue tikus Rattus Norvegicus Strain Wistar Jantan yang diberi diet aterogenik modifikasi standar AIN-93M dan diberi diet normal standar AIN-93M.
English Abstract
Atherosclerosis is a complex chronic disease characterized by the accumulation of lipids within arterial walls which create plaques and can causes constriction, ossification, and/or complete blockage of arteries. BAT is a tissue which is rich of mitocondria that has a role in lowering storage of fat adiposa by means of PPARs α receptor transcription gene and secretion anti inflamation adiponectine. It is able to inhibit plaques of aterosclerosis. Atherogenic diet standard AIN-93M modification as a high fat diet modified from standard diet AIN-93M with the addition of cholic acid and cholesterol is necessarily needed to establish an atherogenic condition. This study aimed to determine the differences between the administration of atherogenic diet standard AIN-93M modification with a normal standard diet of AIN-93M to the amount of Brown Adipose Tissue of male Rattus Norvegicus Strain Wistar rats. This study used a true experimental laboratoric with control group post test design using male Rattus Norvegicus Strain Wistar rats. Sampling was carried out by completely random sampling with 32 rats for total sample. The result tested statistically using independent T-test. The result showed, that atherogenic AIN group (P1) with an average intake of 8,6 ± 2,01 g, energy 37,1 ± 8,6 kcal, and total of Brown Adipose Tissue 0,15 ± 0,27g, meanwhile Normal AIN (P2) with an average intake of10,6 ± 1,27 g, energy 35,6 ± 4,26 kcal, and total of Brown Adipose Tissue 0,12±0,35 g. The raising of BAT in aterogenic diet (P1) is expected as body homeostasis mechanism toward dyslipidemia without obesity especially hypercholesterolemia condition to decrease the lipid acumulation in adipose tissue and lipid toxicity to avoid obesity. Based on this study, it can be concluded that there are significant differences in the amount of Brown Adipose Tissue in rats which is fed by atherogenic diet standard AIN – 93M modification and normal diet standard AIN – 93M.
