Abstract

Latar Belakang: Penderita diabetes mellitus tipe II diketahui memiliki jumlah foam cell yang lebih tinggi dibandingkan dengan orang normal. Jumlah foam cell dapat mengindikasikan risiko kerusakan endotelial pembuluh darah terkait komplikasi diabetes mellitus tipe II. Susu sapi bubuk mengandung vitamin D dan kalsium yang dapat menekan pembentukan foam cell dengan mencegah penyerapan LDL teroksidasi serta menurunkan kadar LDL dan trigliserida pada penderita diabetes mellitus tipe II. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian susu sapi bubuk terhadap jumlah foam cell intima aorta pada tikus putih jantan model diabetes mellitus tipe II yang diberikan diet tinggi lemak dan induksi Streptozotocin. Metode: Penelitian eksperimental menggunakan Post Test Only Control Group Design. Sejumlah 30 tikus wistar dibagi secara acak menjadi 5 kelompok. Semua tikus wistar diadaptasi 1 minggu dengan diet normal, dilanjutkan diet tinggi lemak selama 4 minggu. Pemberian susu sapi bubuk dilakukan selama 56 hari menggunakan susu sapi bubuk komersial dengan mencampurkan ke dalam pakan tikus. Pada hari ke-96 tikus dibedah dan diambil jaringan aortanya, kemudian dibuat preparat histopatologisnya dengan pewarnaan Hematoksilin-Eosin. Selanjutnya dilakukan pembacaan preparat dan penghitungan foam cell dengan aplikasi OlyVIA. Analisa statistik dilakukan dengan menggunakan uji One-Way ANOVA dan uji Korelasi-Regresi Spearman. Hasil: Terdapat perbedaan jumlah foam cell antar kelompok dengan p = 0,000 (p < 0,05) dan terdapat pengaruh pemberian susu sapi bubuk dengan berbagai dosis terhadap jumlah foam cell intima aorta tikus dengan p = 0,009 (p < 0,05). Kesimpulan: Pemberian susu sapi bubuk selama 8 minggu mampu mencegah peningkatan jumlah foam cell intima aorta pada tikus dengan diabetes mellitus secara signifikan dan dosis yang paling efektif adalah dosis 1,8 gram susu sapi bubuk perhari.

English Abstract

Background: The number of foam cells is increased in patients with type 2 diabetes mellitus than normal individuals. Vascular impairments related to type 2 diabetes complications may be indicated from the number of foam cells. Cow powdered-milk containing vitamin D and calcium that can suppress the formation of foam cells by preventing the absorption of oxidized-LDL and reducing LDL and triglyceride levels in patients with type 2 diabetes mellitus. This study was performed to investigate the effect of cow powdered-milk to the number of foam cells in rats with type 2 diabetes mellitus induced with Streptozotocin and high fat diet. Methods: This was an experimental study using Post Test Only Control Group Design. Thirty Wistar rats were randomly divided into 5 groups. All of Wistar rats were adapted to a normal diet in 1 week, then the groups K (+), P1, P2, and P3 were given a high fat diet for 4 weeks. Commercial product of cow powdered-milk was used in this study and given together with rats’ food in 56 days. On 96th day rat tissues dissected and the aorta were taken, then made histopathologic preparations with Hematoxylin-Eosin staining. Further, the number of aortic intima foam cells were quantified by OlyVIA applications. Statistical analysis was performed using One-Way ANOVA and Spearman correlation test. Results: There were differences in the number of foam cells between groups (p = 0,000; p < 0,05) and there was an effect of cow powdered-milk with various doses in the number of rat aortic intima foam cells (p = 0,009; p < 0,05). Conclusion: Giving cow powdered-milk for 8 weeks was able to prevent the increasement on the number of foam cells in aortic intima in rats with diabetes mellitus significantly and the most effective dose is 1.8 g / day.
