Abstract

Penyakit kardiovaskular masih menjadi penyebab kematian terbesar di seluruh dunia. Lebih dari 17 juta orang meninggal karena penyakit kardiovaskular di tahun 2008. Sejumlah 7,3 juta orang meninggal karena sakit jantung iskemik atau penyakit arteri koroner. Antioksidan seperti polifenol menjadi salah satu metode mencegah aterosklerosis. Salah satu tanaman yang mengandung kandungan tinggi polifenol adalah rhizoma rumput teki (Cyperus rotundus). Penelitian ini bertujuan untuk membuktikan bahwa pemberian ekstrak umbi rumput teki berbagai dosis dapat menghambat peningkatan TNF-α pada tikus Rattus norvegicus strain wistar dengan diet aterogenik. Studi ini menggunakan metode randomized post test only controlled group design. Sampel dipilih secara acak kemudian dibagi menjadi 5 kelompok dengan 5 pengulangan yakni kelompok kontrol negatif (diet normal), kelompok kontrol positif (diet aterogenik), dan kelompok diet aterogenik dengan pemberian ekstrak etanol dari umbi rumput teki 6.14 mg/hari, 12.28 mg/hari, 24.56 mg/hari sebagai kelompok perlakuan 1, 2, dan 3. Pemberian diet dan ekstrak dilakukan selama 60 hari kemudian hewan coba dikorbankan untuk diambil darahnya dari jantung. Hasil perhitungan TNF-α serum dengan metode ELISA menunjukkan bahwa pada kelompok tikus kontrol positif dengan kelompok tikus yang diberi diet aterogenik dengan ekstrak etanol umbi rumput teki menunjukan penurunan kadar TNF-α yang bermakna, namun pada setiap peningkatan dosis ekstrak umbi rumput teki (P1 = 57,89 ± 11,6889 ng/mL, P2 = 57,72 ± 13,77913 ng/mL dan P3 = 57,72 ± 13,77913 ng/mL) tidak terdapat penurunan kadar TNF-α yang bermakna. Dari hasil penelitian ini, terdapat hubungan negatif antara peningkatan dosis ekstrak etanol umbi rumput teki dengan kadar TNF-α (Uji Korelasi Pearson R = -0,691, p<0,05). Berdasarkan hasil penelitian ini, disimpulkan bahwa pemberian ekstrak rhizoma rumput teki dapat menghambat peningkatan kadar TNF-α serum secara signifikan pada tikus Wistar yang diberi diet aterogenik. Sehingga perlu dilakukan penelitian lebih lanjut mengenai dosis yang efektif untuk menurunkan kadar TNF- α.

English Abstract

Cardiovascular diseases (CVDs) remain the biggest cause of deaths worldwide. More than 17 million people died from CVDs in 2008. Ischemic heart diseases or coronary heart disease were responsible for 7,3 million deaths. Antioxidant such as polyphenol is one of atherosclerosis prevention methods. One of the plants that contain high level of polyphenol are nut grass (Cyperus rotundus) rhizome. The purpose of this study was to prove that various doses of nut grass rhizome extract could inhibihit TNF-α serum enhancement in wistar rats feed with atherogenic diet. This experimental study used randomized post test only controlled group design. The subjects was randomly selected and divided into five groups with 5 samples each groups that was negative control group (normal diet), positive control group (atherogenic diet), and treatment groups that given atherogenic diet added with 6.14 mg/day; 12.28 mg/day; and 24.56 mg/day nut grass rhizome extract as treatment group 1, 2, and 3. Diet and extract administered for 60 days and then sacrificed to take blood from the heart. The result of TNF-α serum calculated with ELISA method indicated that in negative control group with wistar rats given atherogenic diet with various dose of nut grass rhizome extract show significantly decreasing level of TNF-α, however there was unsignificant decreasing of level of TNF-α among treatment group (P1 = 57,89 ± 11,6889 ng/mL, P2 = 57,72 ± 13,77913 ng/mL dan P3 = 57,72 ± 13,77913 ng/mL). There was negative correlation between increasing dose of nut grass rhizome extract with level of TNF-α (Pearson correlation test R = -0.691, p<0.05). It is concluded that nut grass (Cyperus rotundus) rhizome extract could significantly inhibihit TNF-α serum level enhancement of wistar rats feed with atherogenic diet. So it is necessary to conduct further research regarding the effective dose to decreasing levels of TNF-α.
