Abstract

Hsp27 Terekspresi Secara Berlebihan Pada Kanker Serviks Sebagai Respon Terhadap Kondisi Stres. Ekspresi Hsp27 Yang Berlebihan Dapat Meningkatkan Jalur Invasi, Migrasi, Adhesi Sel Kanker Sehingga Sel Kanker Berpotensi Untuk Ber Metastase. Supernatan Yoghurt Memiliki Kandungan Scfa Meliputi Asetat, Laktat, Dan Butirat Yang Memiliki Aktivitas Antikanker. Tujuan Penelitian Ini Adalah Untuk Mengetahui Dosis Efektif Supernatan Yogurt Lba-St (Lactobacillus Bulgaricus-Acidophilus, Streptococcus Thermophillus) Yang Dapat Menurunkan Jumlah Sel Ekspresi Hsp27. Aktivitas Supernatan Yogurt Dalam Penghambatan Invasi, Migrasi, Dan Adhesi Melalui Imunositokimia. Data Kemudian Dikumpulkan Dan Dianalisis Menggunakan One-Way Anova. Dari Penelitian Ini Dapat Disimpulkan Bahwa Ekspresi Protein Yang Berperan Pada Invasi, Adhesi, Dan Migrasi Yaitu Hsp27 Terbukti Dapat Diturunkan Dengan Dosis Efektif Supernatant Yoghurt Sebesar 20% (V/V) Menyebabkan Penurunan Paling Signifikan Terhadap Jumlah Sel Dan Ekspresi Heat Shock Protein 27 (Hsp27) Terhadap Sel Hela (Cell Line Kanker Serviks) (P <0.05 ± 0.005).

English Abstract

Heat Shock Protein 27 (Hsp27) Is Overexpressed In Cervical Cancer As A Response To Stress Conditions. Hsp27 Overexpression Increase Invasion, Migration, Adhesion Pathways Of Cancer Cells. The Supernatant Yogurt Contains Short-Chain Fatty Acids (Scfa) Include Acetate, Lactate, And Butyrate Which Have Anticancer Activity. This Study Aim To Investigate Effective Dose Of Supernatant Yoghurt Lba-St (Lactobacillus Bulgaricus-Acidophilus, Streptococcus Thermophillus) Can Decrease The Expression Of Hsp27 In Hela Culture Cells. The Mechanism On How Supernatant Yogurt Potency In The Inhibition Of Invasion, Migration, And Adhesion Through Immunocytochemistry. The Data Is Then Collected And Analyzed Using One-Way Anova. From This Study It Can Be Concluded That The Expression Of Proteins That Play A Role In Invasion, Adhesion, And Migration Of The Hsp27 Proved To Be Decreased And The Effective Dose Yoghurt Supernatant By 20% Can Lead To The Most Significant Decrease Of The Number Of Cells And The Expression Of Heat Shock Protein 27 (Hsp27) On Hela Cells (Cervical Cancer Cell Line) (P <0.05 ± 0.005).
