Abstract

Penyakit kardiovaskular merupakan penyebab terbesar kematian di seluruh dunia. Aterosklerosis merupakan salah satu penyebab yang berperan penting dalam penyakit jantung koroner dan penyakit pembuntuan pembuluh darah yang lain. Proses inflamasi yang dipicu sitokin TNF-α merupakan salah satu proses yang berperan dalam aterogenesis. Ekstrak kulit manggis diketahui memilki efek antiinflamasi dengan menghambat aktivasi NF-κB. Penelitian ini bertujuan untuk membuktikan bahwa ekstrak kulit manggis (Garcinia mangostana L.) dapat menurunkan kadar TNF-α serum pada tikus wistar model aterogenik. Penelitian ini merupakan studi eksperimental dengan post test control group design dengan hewan coba tikus Rattus norvegicus jantan galur Wistar. Sampel dipilih dengan cara proporsional sampling, dibagi dalam sembilan kelompok, yaitu kelompok diet tinggi lemak empat minggu, diet tinggi lemak 12 minggu, diet normal, diet tinggi lemak selama 12 minggu dan diberi asupan ekstrak kulit manggis dengan dosis 200 mg/kgBB/hari, 400 mg/kgBB/hari dan 800 mg/kgBB/hari serta diet tinggi lemak selama 12 minggu dan pemberian ekstrak kulit manggis dengan dosis 200 mg/kgBB/hari, 400 mg/kgBB/hari, dan 800 mg/kgBB/hari selama delapan minggu (dimulai empat minggu setelah pemberian diet tinggi lemak). Variabel yang diukur adalah kadar TNF-α serum tikus yang dipapar diet aterogenik. Hasil penelitian menunjukkan terjadi penurunan kadar TNF-α pada kelompok tikus yang diberi ekstrak kulit manggis berbagai dosis berbeda bermakna (Anova, p<0,05). Terbukti adanya perbedaan perubahan kadar TNF-α antar kelompok tikus yang diberi ekstrak kulit manggis baik selama 8 minggu maupun 12 minggu (Uji Post Hoc Tukey, p<0,05). Hasil uji korelasi Pearson sebesar -0,942 pada pemberian ekstrak kulit manggis selama 12 minggu dan -0,690 pada pemberian ekstrak kulit manggis selama 8 minggu menunjukkan korelasi yang kuat antara peningkatan dosis ekstrak dan penurunan kadar TNF-α. Kesimpulan dari penelitian ini, ekstrak kulit manggis dapat secara signifikan menghambat peningkatan dan menurunkan kadar TNF-α tikus yang diberi diet tinggi lemak

English Abstract

Cardiovascular disease is one of the most cause of death in the world. Atherosclerosis is one of the causes that is important for cardiovascular disease. Inflammation which is stimulated by TNF-α is on of the process which is contributed in atherogenesis. Mangosteen pericarp extract is known has anti-inflammatory effect that can inhibit activation of NF-κB. The goal of this study is to prove anti-inflammatory effect from mangosteen pericarp extract by decreasing TNF-α serum in rat with high fat diet. This study is an experimental study with post-test control group design with Rattus norvegicus Wistar male rats. The sample was selected by means of proportional sampling, were divided into nine groups, namely the four-week high-fat diet, high-fat diet 12 weeks, a normal diet, high-fat diet for 12 weeks and fed a diet of mangosteen pericarp extract at a dose of 200 mg / kg / day, 400 mg / kg / day and 800 mg / kg / day as well as a high-fat diet for 12 weeks and mangosteen pericarp extract at a dose of 200 mg / kg / day, 400 mg / kg / day, and 800 mg / kg / day for eight week (starting four weeks after administration of a high-fat diet). The variables measured were serum levels of TNF-α rats were exposed to high fat diet. The results showed decreased levels of TNF-α in mice fed various doses of mangosteen peel extract differ significantly (ANOVA, p<0.05). There is evidence of differences in the levels of TNF-α changes in between mice fed the extract (Post Hoc Tukey test, p<0.05). The results of Pearson correlation test for magosteen pericarp administration for 12 weeks is -0.942 and for 8 weeks is -0.690 showed that increasing doses of the extract make decreased levels of TNF-α. The conclusion of this study, mangosteen pericarp extract can significantly inhibit increasing and reduce levels of TNF-α rat with high-fat diet.
