Abstract
Salah satu prinsip penelitian hewan coba adalah refinement, memberikan hewan coba akses makanan dan minuman yang sesuai kebutuhannya. Sehingga dibutuhkan penerapan diet standar untuk hewan coba. Umumnya Diet standar yang diberikan pada Rattus norvegicus strain wistar ada dua macam yaitu diet normal standar PAR-S dan diet normal standar AIN-93M. Kedua diet tersebut memiliki kandungan zat gizi yang berbeda. Penelitian ini bertujuan mengetahui perbedaan kadar trigliserida serum pada kelompok diet normal standar PAR-S dan diet normal standar modifikasi AIN-93M. Studi eksperimental menggunakan Randomized control Group postest design dilakukan terhadap tikus Wistar jantan di Labolatorium Biosains Universitas Brawijaya selama 12 minggu. Sampel dipilih dengan cara RAL (Rancangan Acak Lengkap) dibagi dalam dua kelompok dengan jumlah total 32 tikus wistar. Variabel yang diukur pada penelitian ini adalah kadar trigliserida serum tikus. Hasil penelitian ini menunjukkan bahwa rata-rata asupan kelompok diet normal standar PAR-S 41,56 ± 5,53 g/hari, sedangkan diet normal standar AIN-93M 10,41 ± 1,18 g/hari. Hasil kadar trigliserida serum berbeda signifikan pada kedua kelompok (uji T, p<0,05), rata-rata kadar trigliserida serum pada kelompok diet normal standar PAR-S 74,88 ± 34,2 mg/dL, dan rata-rata kadar trigliserida serum pada kelompok diet normal standar AIN-93M 47,44 ± 27,09 mg/dL. Kesimpulan penelitian ini terdapat perbedaan yang signifikan kadar trigliserida serum dikarenakan perbedaan jumlah asupan makanan, sehingga kandungan zat gizi yang diterima masing-masing kelompok berbeda.
English Abstract
A refinement is one of the principles of animal research, which means giving the experimental animal to access food and drink to fit their needs. Therefore, applying standard diets for experimental animal is needed. There are two type standard diets to Rattus norvegicus strain Wistar, namely: 1) Standard normal diet PAR-S, 2) Standard normal diet AIN-93M. These diets have difference nutrition contents. The aim of the study is finding the differences of serum triglyceride levels at standard normal diet PAR-S group and standard normal diet AIN-93M modification group. This experimental is using Randomized control Group posttest design performed in male wistar that is located at Biosains Laboratory of Brawijaya University during twelve weeks. Samples are chosen by random sampling divided into two groups with 32 experimental rats. The variable measured is serum triglyceride level Wistar. The result of this study shows that the average intake of standard normal diet PAR-S group 41,56 ± 5,53 g/day, whereas standard normal diet AIN-93M 10,41 ± 1,18 g/day. The analysis serum triglyceride level shows significant difference between two groups (T test, p<0,05), the average of standard normal diet PAR-S serum triglyceride 74,88 ± 34,2 mg/dL, and the average of serum triglyceride standard normal diet AIN-93M 47,44 ± 27,09 mg/dL. This study reveals that there is significant difference serum triglyceride level at two groups, it is because the average of intake food is differents, so nutrition content accepted also different between those groups
