Abstract

Tujuan : Untuk mengetahui keefektifan polifenol daun kersen untuk menghambat pembentukan plak yang dihasilkan bakteri Streptococcus mutans. Metode: Rancangan penelitian adalah true experimental pre control design only secara in vitro. S.mutans dibiakkan dalam media BHI (Brain Heart Infusion). Daun kersen kering seberat 50 gram dilarutkan dalam metanol dengan metode maserasi menghasilkan 10 ml ekstrak polifenol kersen. Ekstrak dibagi menjadi tiga kelompok dosis, yaitu 0,625 ml, 1,25 ml, dan 1,875 ml. Gigi insisivus pertama rahang atas manusia dipotong enamelnya dijadikan sebagai media in vitro, direndam ke dalam ekstrak yang telah ditambahkan susu yang mengandung sukrosa. Selanjutnya diinkubasi selama 2x24 jam secara anaerob bersama bakteri S.mutans 108/cc. Setelah 2x24 jam, dialirkan NaCl 0,9% untuk meluruhkan bagian yang tidak adheren dan dicuci dengan aquadest. Tahap akhir, diaplikasikan bahan disclosing agent untuk mendeteksi plak pada permukaan enamel. Parameter yang diukur adalah Rasio antara luas plak (area terwarna) dan luas permukaan email gigi. Hasil: Rasio ekstrak daun kersen pada kelompok kontrol negatif adalah 0,69%, kontrol positif 0,61%, dosis 0,625 ml adalah 0,21%, pada dosis 1,25 mg adalah 0,53%, dan dosis 1,875 ml adalah 0,53%. Semakin tinggi rasio ekstrak, semakin banyak pembentukan plak. Setelah diuji statistik one way ANOVA, dosis 0,625 ml berbeda bermakna (p < 0,05) dengan dosis yang lain. Kesimpulan: Polifenol daun kersen dapat menghambat pembentukan plak bakteri S.mutans, dengan dosis yang paling efektif yaitu 0,625 ml.

English Abstract

Objective: To determine the effectiveness of Muntingia calabura leaves polyphenols to inhibit the formation of plaque bacteria Streptococcus mutans produced on dental caries prevention. Methods: The study design is true experimental design pre-control only in vitro. Streptococcus mutans was cultured in BHI (Brain Heart Infusion) medium. Dried leaves weighing 50 grams was dissolved in methanol by maceration method resulted in 10 ml Muntingia calabura polyphenol extract. Extracts were divided into three dose groups, namely 0,625 ml, 1,25 ml, and 1,875 ml. Enamel of maxillary first incisors man were cut and used as an in vitro medium, soaked in extracts and added milk that containing sucrose. Furthermore, incubated anaerobically for 2x24 hours with S.mutans 108/cc. After that, flowed NaCl 0,9% to remove any non-adherent bacteria and washed with distilled water. The final stage, disclosing agent was applied to detect plaque on the enamel surface. The parameter is ratio of area measured plaque (coloured area) and surface area of tooth enamel. Results: Ratio of Muntingia calabura leaves extract in negative control group was 0,69%, positive control was 0,61%, 0,625 ml dose was 0,21%, 1,25 ml dose was 0,53%, and 1,875 ml dose was 0,35%. The higher ratio of the extract is the more plaque formation. After one-way ANOVA statistical test, a dose of 0,625 mg was significantly different (p < 0.05) with another dose. Conclusion: Muntingia calabura leaves polyphenols can inhibit the formation of plaque bacteria Streptococcus mutans, the most effective dose is 0,625 ml
