Abstract
Melalui pemeriksaan biopsi hati, dapat diketahui derajat fibrosis hati pada pasien. Tindakan invasif ini memiliki banyak komplikasi, seperti nyeri, pneumothorax, hemothorax, dan pendarahan yang dapat menyebabkan kematian. Penelitian ini bertujuan untuk melihat apakah pemeriksaan kadar IL-17 serum dapat menggantikan biopsi hati. Dalam penelitian ini, dilihat perbandingan ekspresi IL-17 di jaringan dan kadar IL-17 di serum dengan derajat fibrosis hati pada tikus yang diinduksi Karbon Tetraklorida (CCl4): Sampel yang digunakan adalah 24 ekor tikus Rattus novergicus galur Wistar yang dibagi dalam 4 kelompok dengan derajat fibrosis yang berbeda, kelompok kontrol (derajat 0), kelompok perlakuan dengan pemberian CCl4 1 mL/KgBB setiap 72 jam selama 2 minggu untuk derajat 1, 5 minggu untuk derajat 2, dan 9 minggu derajat 3. Ekspresi IL-17 jaringan diperiksa dengan pengecatan imunohistokimia dan kadar IL-17 serum dengan ELISA yang kemudian dibandingkan pada tiap derajat fibrosis. Didapatkan kesimpulan bahwa ekspresi dan kadar IL-17 memiliki hubungan korelasi yang lemah (r = 0.486), dan sebanding hanya pada derajat 3 fibrosis, sehingga dengan kata lain pemeriksaan kadar IL-17 serum dapat mewakili derajat fibrosis yang dialami hanya pada derajat 3.
English Abstract
Until now the determination of liver fibrosis stage is performed using liver biopsy. This invasive technique causes many complications, like severe pain, pneumothorax, hemothorax, and massive bleeding that may lead to death. This research aims to observe whether liver biopsy can be replaced by measurement of IL-17 serum level in liver fibrosis screening. In this research, we studied the comparison of IL-17 expression in liver and its serum level with the stages of liver fibrosis in rats induced by carbon tetrachloride: The samples were 24 Rattus novergicus strain Wistar, divided into 4 groups with different fibrosis stage, which are control group (stage 0), treatment group treated with CCl4 1 mL/KgBB every 72 hours for 2 ,5, and 9 weeks to reach stage 1, 2, and 3 subsequently. Immunohistochemistry staining and ELISA is done to assess IL-17 expression in liver and IL-17 serum level, which then compared in every fibrosis stage. It is concluded that IL-17 expression and serum level have a moderate correlation (r = 0.468) and are comparable only at third stage of fibrosis, so, in other words, IL-17 serum level checking can represent fibrosis stage only at the third stage.
