Abstract

Atherosklerosis merupakan proses inflamasi pada pembuluh darah yang disertai aktivitas imun. Gambaran histopatologi berupa peningkatan ketebalan aorta. Manifestasi klinis yang ditimbulkan mulai dari penyakit kardio-vaskuler hingga gangguan organ-organ lain. Terapi dengan statin belum efektif menurunkan progresifitas lesi karena statin hanya bekerja pada salah satu factor risiko, yaitu menurunkan kadar kolesterol dalam darah. HSP60 merupakan protein protektif di dinding arteri manusia dan terekspresi saat terdapat stress endotel. HSP65 merupakan protein homolog HSP60 yang terdapat pada bakteri, seperti Mycobacterium tuberculosis. Tujuan penelitian ini adalah mengetahui pengaruh pemberian vaksinasi HSP65 pada aorta mencit yang telah diinduksi atherosklerosis melalui mekanisme toleransi mukosa. Penelitian ini merupakan penelitian eksperimental menggunakan post test control group design terbagi dalam 5 kelompok dengan masing-masing terdiri dari 6 mencit. Kelompok I mencit diberi diet normokolesterolemia (kontrol negatif), kelompok II mencit diberi diet atherogenik tanpa vaksinasi (kontrol positif), kelompok III mencit diberi diet atherogenik dan vaksinasi 0,5μg, kelompok IV mencit diberi diet atherogenik dan vaksinasi 0,75μg, sedangkan kelompok V mencit diberi diet atherogenik dan vaksinasi 1μg. Hasil pengukuran ketebalan aorta dengan pengecatan hematoxylin eosin, berbeda secara bermakna antara mencit kontrol positif dengan mencit perlakuan vaksinasi (ANOVA, p<0,05) sedangkan ketebalan aorta hampir sama antara mencit kontrol negatif dengan mencit perlakuan vaksinasi dosis II (ANOVA, p>0,05). Melalui analisis korelasi Pearson didapatkan hubungan yang bermakna dengan angka signifikansi 0,024 dan kekuatan korelasi sebesar -0,449. Kesimpulan penelitian ini adalah vaksinasi dengan HSP65 Mycobacterium tuberculosis dapat menurunkan ketebalan aorta pada mencit model atherosklerosis dan dosis efektifnya adalah 0,75 μg.

English Abstract

Atherosclerosis is an inflammatory process of the blood vessels accompanied by immune activity. Histopathologic features in the form of an increase in the thickness of the aortic lumen. Clinical manifestations arising from cardio-vascular disease to disease affecting other organs. Treatment with statins have not been effective in lowering the progression of atherosclerotic lesions as statins only work on one risk factor for atherosclerosis, which lowers blood cholesterol levels. HSP60 is a protective protein in the human artery wall and endothelial-expressed when there is stress. HSP65 is homologous protein HSP60 found in bacteria, such as Mycobacterium tuberculosis. The purpose of this study was to determine the effect of HSP65 on the vaccination through mucosal tolerance of mice that have been induced aortic atherosclerosis. This study was an experimental study using post-test control group design in which subjects were divided into 5 groups. Each group consisted of 5 mice. Group I mice fed a diet normokolesterolemia (negative control), group II mice given atherogenic diet without vaccine (positive control), group III mice given atherogenic diet and 0,5μg accination, group IV mice were given atherogenic diet and 0,75μg vaccination, group V mice were given atherogenic diet and 1μg vaccination. The results of the thickness measurement of the aorta with hematoxylin eosin staining, was significantly different between the positive control mice treated mice vaccinated (ANOVA, p<0.05), while the thickness of the aorta is almost the same between the negative control mice treated mice vaccinated with the second dose (ANOVA, p>0.05). Through Pearson correlations analytic there is a significant relation with signification 0,024. The conclusion of this study is vaccination with Mycobacterium tuberculosis HSP65 can reduce the thickness of the aorta in mice induced atherosclerosis and the effective dose is 0.75 mg dose.
