Abstract

Perlemakan hati merupakan akumulasi lemak yang berlebih di dalam sel hati melebihi 5% dari berat organ hati sebagai akibat tingginya inflamasi yang progresif. Inflamasi tersebut dikarenakan peningkatan pembentukan Reactive Oxygen Species (ROS). ROS akan mengaktifkan NF-κβ sehingga memicu upregulasi sitokin TNF-α. ROS dapat dicegah dengan fitosterol dan flavonoid. Salah satu tanaman yang memiliki kandungan fitosterol dan flavonoid tinggi yaitu daun katuk (Sauropus adrogynus L. Merr). Penelitian ini bertujuan mengetahui pengaruh pemberian bubuk daun katuk berbagai dosis terhadap penurunan kadar TNF-α jaringan hepar tikus yang diberi diet aterogenik. Studi eksperimental ini menggunakan Post Test Control Group Design, dilakukan pada 25 ekor tikus Wistar jantan yang dibagi secara acak menjadi 5 kelompok. Kelompok pertama K(-)/kontrol negatif adalah tikus dengan diet normal, kelompok ke-2 K(+)/kontrol positif diberi diet aterogenik, dan kelompok ke-3 sampai ke-5 (P1, P2 dan P3) diberi diet aterogenik dengan subtitusi bubuk daun katuk 6%, 9% dan 12%. Pemberian bubuk daun katuk diberikan secara per oral (ad libitum) setiap hari selama 60 hari. Parameter yang diukur adalah kadar TNF-α jaringan hepar. Hasil penelitian menunjukkan bahwa rata-rata kadar TNF-α tertinggi diperoleh pada kelompok K(+) yaitu 291,28 ± 79,30 ng/mL dan kadar TNF-α terendah di antara kelompok perlakukan diperoleh kelompok P3 yaitu 184,9 ± 33,74 ng/mL. Pemberian bubuk daun katuk dapat menurunkan kadar TNF-α secara signifikan p = 0,01 (p < 0,05). Disimpulkan bahwa pemberian bubuk daun katuk memberikan efek yang signifikan dalam menurunkan kadar TNF-α jaringan hepar.

English Abstract

Fatty liver is an accumulation of excess fat in the liver cells exceeds 5% of the entire weight of the liver as a result of high progressive inflammation. Inflammation is due to increased formation of Reactive Oxygen Species (ROS). ROS activates NF-κβ triggering upregulation of cytokines TNF-α. ROS can be prevented with phytosterols and flavonoid. One of the plants that contain high level of phytosterol and flavonoid are katuk leaf (Sauropus adrogynus L. Merr). The objective of study was to find out the effect of katuk leaf powder of different dosages on Liver Tissue Levels of TNF-α of rats with atherogenic diet. The Experimental study was using Post Test Control Group Design, conducted on 25 male Wistar rats were divided randomly into 5 groups. First group K(-) was given normal diet (negative control), second group K(+) was given atherogenic diet (positive control), and group three to five (P1, P2, and P3) was given atherogenic diet with katuk leaf powder substitution 6%, 9% and 12%. Katuk leaf powder was given orally (ad libitum) in 60 days. The parameter has been measured was liver tissue levels of TNF-α. The result showed that the highest level of TNF-α were observed for K(+) group were 291,28 ±79,30 ng/mL and the lowest level of TNF-α in treatment group were observed for P3 was 184,9 ± 33, ng/mL. The giving of katuk leaf powder were able to decrease the amount of significantly TNF-α level p = 0,01 (p < 0,05). It can be conclude that giving katuk leaf powder have significant effect for decrease TNF-α level.
