Abstract

Diabetes Mellitus (DM) merupakan penyakit gangguan metabolisme yang dapat disebabkan karena penurunan kemampuan sel beta pankreas untuk mensekresi insulin sebagai respon terhadap beban glukosa. Untuk mengatasi hal tersebut telah dilakukan pendekatan terapi makanan berbasis alam. Jamur tiram putih memiliki kandungan beta-glucan (β-Glucan) yang tinggi dimana pada mencit terbukti meningkatkan Granulocyte-Colony Stimulating Factor (G-CSF), sehingga dapat memperbaiki jaringan organ yang rusak, termasuk sel beta pankreas. Penelitian ini dilakukan untuk membuktikan pengaruh pemberian snack dari jamur tiram putih (Pleorotus ostreatus) stick (Pleostick) terhadap kadar glukosa darah, pada tikus hasil induksi Multiple Low Dose-Streptozotocin (MLD-STZ) dosis 35 mg/kgBB. Penelitian ini menggunakan rancangan acak lengkap dengan Post test Control Group Design, dilakukan pada 30 ekor tikus Wistar jantan yang dibagi secara acak menjadi 5 kelompok. Kelompok pertama kontrol negatif (KN) adalah tikus dengan diet normal, kelompok ke-2 kontrol positif (KP) diberi diet tinggi lemak (High Fat Diet), dan kelompok ke-3 sampai ke-5 (P1, P2 dan P3) diberi Pleostick sebesar 7,5 g, 15 g, dan 30 g dengan dosis β-Glucan sebesar 100mg/kgBB, 200mg/kgBB dan 400mg/kgBB per oral setiap hari selama 30 hari. Kadar glukosa darah tikus diukur pada awal, setelah 2 bulan, dan akhir penelitian. Uji statistik yang digunakan One Way ANOVA dan Kruskall Wallis (p<0,05). Pemberian Pleostick pada tikus model DM Tipe 2 menunjukkan perubahan rata-rata kadar glukosa darah (p=0,036). Pemberian Pleostick menurunkan kadar glukosa darah secara signifikan (p=0,008) pada kelompok P2. Penelitian ini menunjukkan pemberian snack dari jamur tiram putih (Pleostick) terbukti mampu menurunkan kadar glukosa darah tikus.

English Abstract

Diabetes Mellitus (DM) is a metabolic disorder that can be caused due to a decrease in the ability of pancreatic beta cells to secrete insulin in response to glucose load. This therapeutic approach has been taken based natural foods. White oyster mushroom contains of high beta-glucan (β-Glucan) which is shown to increase Granulocyte-Colony Stimulating Factor (G-CSF) on diabetes melitus mice, so it can repair damaged tissue, including pancreatic beta cells. This study was conducted to prove the effect of giving a snack based on white oyster mushroom (Pleorotus ostreatus) stick (Pleostick) on blood glucose levels, in mice induce Multiple Low-Dose Streptozotocin (MLD-STZ) of 35 mg/kg. This study used a completely randomized design with post test control group design, conducted on 30 male Wistar rats were divided randomly into 5 groups. The first group of negative control (KN) is rats with normal diet, group-2 to the positive control (KP) were given a high-fat diet (High Fat Diet), and the group to 3-to-5 (P1, P2 and P3) were given 7.5 g, 15 g, and 30 g Pleostick that equal to 100 mg/kg, 200 mg/kg and 400 mg/kg of β-Glucan orally daily for 30 days. Mice blood glucose levels were measured at baseline, 2 months after induce STZ, and 1 month after STZ and Pleostick intervention. The analysis is One Way ANOVA and Kruskall Wallis (p <0.05). Giving Pleostick in mice model of type 2 diabetes showed an average change in blood glucose levels (p = 0.036). Giving Pleostick lowers blood glucose levels were significantly (p = 0.008) in the group of P2. This study shows giving snack based on white oyster mushroom (Pleostick) proved capable of lowering blood glucose levels of mice.
