Abstract
Latar Belakang : Pada Dm2 Terjadi Resistensi Insulin Yang Ditandai Hiperinsulinemia. Binahong Merupakan Tanaman Yang Digunakan Dalam Pengobatan Dm2 Secara Tradisional. Tujuan: Mengetahui Efek Ekstrak Daun Binahong Terhadap Kadar Glukosa Darah Dan Serum Insulin. Metode : True Experimental In Vivo , Dengan Sampel Tikus Wistar Berjumlah 30 Ekor. Sampel Dibagi Ke Dalam 6 Kelompok Perlakuan Yaitu (I) Kelompok Normal, (Ii) Kontrol Dm2, (Iii) Kontrol Pembanding (Dm2 Dengan Glimepiride), Dan (Iv-Vi) 3 Kelompok Perlakuan (Dm2 Dengan Ekstrak Daun Binahong Dosis 17,5mg/Kgbb; 35 Mg/Kgbb; Atau 70 Mg/Kgbb) Diberikan Per Hari Secara Oral Selama 15 Hari. Induksi Dm2 Dilakukan Dengan Pemberian Diet Tinggi Lemak Selama Lima Minggu Dan Injeksi Tunggal Stz (35 Mg/Kgbb) Secara Intraperitonial. Kadar Glukosa Darah Diamati Pada Tiap Minggu, Sedangkan Kadar Serum Insulin Diukur Menggunakan Elisa Kits. Hasil : Semua Kelompok Perlakuan Menunjukkan Perbedaan Penurunan Glukosa Darah, Kadar Serum Insulin, Dan Homa-Ir Yang Bermakna (P<0,05) Dibandingkan Kontrol Diabetes. Dosis 35 Mg/Kgbb Mencapai Kadar Glukosa Darah Normal (< 200 Mg/Dl). Dosis 35 Mg/Kgbb Tidak Menunjukkan Perbedaan Bermakna Pada Kadar Serum Insulin (P=0,180) Dan Homa-Ir (P=0,297) Dibandingkan Kontrol Normal. Kesimpulan : Ekstrak Daun Binahong Mampu Menurunkan Kadar Glukosa Darah, Serum Insulin Dan Resistensi Insulin. Dosis 35 Mg/Kgbb Merupakan Dosis Optimal.
English Abstract
Introduction: Dm2 Is Characterized By Insulin Resistance With Hyperinsulinemia. Binahong Leaves Are Traditionally Used To Treat Dm2. Objectives: To Examine The Effect Of Binahong Leaves To Decrease Blood Glucose And Insulin Serum Levels. Methods : True Experimental In Vivo , Which Thirty Male Wistar Rats Were Randomly Assigned Into 6 Groups: (I) Normal Control; (Ii) Dm2 Control; (Iii) Control Standard Drug (Dm2 And Glimepiride); (Iv-Vi) 3 Treatment Groups (Dm2 And Binahong Leaves Extract 17,5 Mg/Kg.Bw; 35 Mg/Kg.Bw; Or 70 Mg/Kg.Bw) Were Administrated To The Diabetic Rats Per Day Orally For 15 Days. Dm2 Was Induced In Rats By Fed High Fat Diet In 5 Weeks And Intraperitoneally (I.P.) Injection Of Stz (35 Mg/Kg.Bw). Blood Glucose Levels Were Observed Every Week And Insulin Serum Level Was Measured With Elisa Kits. Results: All Of Treatment Groups Showed Significantly Decrease Of Blood Glucose Levels, Insulin Serum, And Homa-Ir (P<0,05) Compared To Dm2 Control. Dose Of 35 Mg/Kg.Bw Produced No Significant Difference In Insulin Serum Levels (P=0,180) And Homa-Ir (P=0,297) Compared To Normal Control. Conclusion: Extract Of Binahong Leaves At A Dose Of 35 Mg/Kg.Bw Can Decrease Blood Glucose, Insulin Serum Levels, And Insulin Resistance. 35 Mg/Kgbb Of Dose Is The Optimum Dose.
