Abstract
Asiatikosida merupakan senyawa marker dalam tanaman pegagan (Centella asiatica) yang mempunyai khasiat obat terutama sebagai antiinflamasi. Asiatikosida memiliki sifat hidrofil dengan molekul yang besar sehingga penetrasi menembus membran sel akan lebih sulit. Oleh karena itu diperlukan modifikasi kepolaran asiatikosida melalui sistem penghantaran obat yaitu fitosom. fitosom yang stabil ditandai dengan adanya morfologi dan ukuran, pH, Entrapment Efficiency (EE), serta kadar senyawa yang dikandungnya tetap stabil selama penyimpanan. Salah satu faktor yang mempengaruhi kestabilan adalah komponen penyusunnya untuk itu penelitian ini bertujuan untuk mengetahui pengaruh penambahan bahan peningkat stabilitas cangkang yaitu kolesterol terhadap stabilitas fisika dan kimia fitosom ekstrak pegagan. Hasil uji stabilitas menunjukan bahwa tidak terdapat perbedaan kestabilan fisika dan kimia fitosom ekstrak pegagan antara formula fitosom tanpa penambahan kolesterol (F1) dan formula fitosom dengan penambahan kolesterol (F2). Namun nilai sifat fisika seperti ukuran pastikel, pH, EE yang dihasilkan oleh formula dengan penambahan kolesterol (F2) lebih besar dibandingkan dengan formula tanpa penambahan kolesterol (F1). Sedangkan untuk parameter kimianya seperti kadar F2 cenderung lebih kecil yang terukur. Bentuk fitosom yang dihasilkan dari F2 yaitu sferik dengan ukuran partikel 1,13- 1,59 μm, entrapment efficiency sebesar ± 85%, pH sebesar ± 5,6 dan kadar asiatikosida sebesar 0,215%.
English Abstract
Asiaticoside is marker compounds in gotu kola extract (Centella asiatica) which has primarily as an anti-inflammatory drug efficacy. Asiaticoside is hydrophilic and large molecule fitoconstituen, because of its characteristic make the penetration more difficult. Therefore polarity modification of Asiaticoside is needed, one of it is by phytosome. Stable phytosome characterized by morphology and size, pH, Entrapment Efficiency (EE), and levels compound remained stable during storage. One of the factors that affect stability are the component of phytosome, this study aims to determine the effect of shell stability enhancer (cholesterol) on the physical and chemical stability phytosome gotu kola extract. The results of the stability test showed that there were no differences in the physical and chemical stability of gotu kola extract phytosome between formula without the addition of cholesterol (F1) and formula with the addition of cholesterol (F2). However, the value of physical properties such as size, pH, EE produced by a formula with the addition of cholesterol (F2) is greater than the formula without the addition of cholesterol (F1). nevertheless for chemical parameters (level of asiaticoside) F2 is smaller than F1. Phytosome Characteristic of F2 was spheric with a particle size 1.13 -1.59 μm, entrapment efficiency ± 85%, pH ± 5.6 and content asiaticoside was 0.215%.
