Abstract
Interleukin-1 b (IL-1 b ) adalah sitokin proinflamasi yang menginduksi kerusakan sel b pankreas pada kondisi diabetes melitus tipe 2. Susu sapi bubuk sebagai sumber vitamin D dapat menurunkan kadar IL-1 b pada sel b pakreas. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian susu sapi bubuk terhadap kadar IL-1 b tikus wistar model DM tipe 2. Penelitian ini merupakan penelitian eksperimental dengan desain post test only control group . Penelitian dilakukan selama 90 hari dengan menggunakan 30 ekor tikus wistar jantan yang dibagi secara acak menjadi 5 kelompok yaitu: P0 (diet normal), P1 ( high fat diet ), P2 ( high fat diet + susu sapi bubuk 0,9 gr), P3 ( high fat diet + susu sapi buuk 1,8 gr), dan P4 ( high fat diet + susu sapi bubuk 2,7 gr). Variabel tergantung adalah kadar IL-1 b yang diukur dengan ELISA kit. Analisis data menggunakan uji One Way ANOVA. Hasil uji One Way ANOVA menunjukkan tidak ada perbedaan yang signifikan kadar IL-1 b diantara kelompok tikus yang diberi susu sapi bubuk p = 0,149 (p>0,05). Hal ini dapat disebabkan oleh beberapa hal, antara lain asupan susu sapi bubuk yang kurang dari dosis, serta kandungan whey protein , SFA, dan TFA dari susu sapi bubuk. Namun, jika dilihat dari grafik, pemberian susu sapi bubuk mampu menurunkan kadar IL-1 b jika dibandingkan dengan kontrol positif. Dosis susu sapi bubuk yang dapat menurunkan kadar IL-1 b tertinggi adalah 2,7 gram/hari. Diperlukan penelitian lanjutan dengan jumlah sampel yang lebih besar dan perlu diperhatikan cara pemberian susu sapi bubuk serta diaplikasikan pada manusia.
English Abstract
Interleukin-1 b (IL-1 b ) is a proinflammatory cytokine that induces pancreas b cell damage in type 2 diabetes mellitus condition. Cow’s milk powder as source of vitamin D can reduce levels of IL-1 b in pancreatic b cells. Purpose of this study is to know the effect of milk powder to the levels of IL-1 b Wistar rat type 2 DM models. Research was an experimental study design with a post-test only control group. The study was conducted for 90 days using 30 male Wistar rats were divided randomly into 5 groups: P0 (normal diet), P1 (high fat diet), P2 (high fat diet + 0,9 g milk powder), P3 ( high fat diet + 1,8 g milk powder), and P4 (high fat diet + 2,7 g milk powder). Dependent variable is the level of IL-1 b which is measured by ELISA kit. Analysis of the data using One Way ANOVA test. The result of One Way ANOVA test showed no significant difference between the levels of IL-1 b mice fed a powdered milk p = 0,149 (p > 0,05). It can be caused by several thing, among others, the intake of cow’s milk powder which is less than the dose, and the amount of whey protein, SFA, and TFA from milk powder. However, when seen from the graph, powdered cow’s milk could reduce levels of IL-1 b when compared with the positive control. Dose of cow’s milk powder which can degrade the highest levels of IL-1 b is 2,7 grams/day. Futher research is needed with a large sample size and to note how cow’s milk powder and applied to human. ��� ;�s���D��=K�nt-family:Arial;mso-char-type:symbol;mso-symbol-font-family: Symbol> b tertinggi adalah 2,7 gram/hari.
