Abstract
Bibir sumbing disebabkan oleh multifaktorial, baik genetik maupun lingkungan. Faktor genetik yang menyebabkan apoptosis dan remodelling pada masa perkembangan janin yang diambil dalam penelitian ini adalah MMP-3 (Matrix Metalloproteinase-3) dan AP-1 (Activating Protein-1). Protein ini memiliki efek yang sinergis dalam proses apoptosis dan remodelling janin. Tujuan penelitian ini adalah untuk mengetahui hubungan antara protein MMP-3 dan AP-1 pada kejadian bibir sumbing dengan menggunakan desain penelitian cross-sectional analytic. Sampel penelitian adalah jaringan bibir sumbing hasil operasi oleh tim bedah plastik rumah sakit Saiful Anwar Malang pada kegiatan bakti sosial tanggal 3,6,7,8 dan 12 Desember 2012 Di RSU Larantuka Kupang, RSU Kupang, dan RSU Alor Nusa Tenggara Timur yang memenuhi kriteria inklusi. Jaringan bibir dilakukan pewarnaan imunohistokimia pada jaringan dengan antibody monoklonal MMP-3 dan antibodi monoklonal AP-1 setelah itu diperiksa dibawah mikroskop dengan menghitung jumlah sel yang mengekspresikan protein tersebut. Hasil analisis korelasi dengan uji Pearson antara protein MMP-3 dan AP-1 menunjukkan hubungan bermakna dengan korelasi antar protein yang cukup (0.429). Kesimpulan dari penelitian ini adalah terdapat hubungan yang positif dan bermakna dengan koefisien korelasi yang cukup kuat antara ekspresi protein MMP-3 dan ekspresi protein AP-1 pada kejadian bibir sumbing ras Protomalayid provinsi Nusa Tenggara Timur.
English Abstract
Cleft lip caused by multifactor, genetic and also environment. Genetic factors which has important role in apoptosis and remodelling in embryo development is the expression of protein MMP-3 (Matrix Metalloproteinase-3) and AP-1 (Activating Protein-1). These proteins have sinergic effect in apoptosis and remodelling of embryo. The aim of this research is to discover the relation of protein MMP-3 and AP-1 in cleft lip cases by using cross-sectional analytic method. The samples of this research were taken on social event by General Hospital of Syaiful Anwar Malang, General Hospital of Alor Nusa Tenggara Timur, and General Hospital of Kupang on December 3rd ,6th ,7th ,8th , and 12th 2012 which fulfill the inclussion criterias. The immunohisto chemistry staining were conducted on each lip tissue by using monoclonal antibody of MMP-3 and monoclonal antibody of AP-1. After that, every tissues were examined under light microscope by counting the cells that expressed those proteins. Corelation analysis of this research by using Pearson test among MMP-3 and AP-1 show positive relation with immediate strength of correlation between the expression of MMP-3 and the expression of AP-1 in cleft lip cases of Protomalayid race, East Nusa Tenggara Province.
