Abstract

Pada diabetes melitus terjadi hiperglikemia yang memicu pembentukan dan peningkatan kadar Reactive Oxygen Species (ROS) sehingga berpotensi menimbulkan stress oksidatif yang ditandai dengan terjadinya peroksidasi lipid. Pengukuran kadar F2-isoprostan dapat digunakan sebagai penanda terjadinya stress oksidatif dalam tubuh penderita DM. Salah satu sumber antioksidan adalah albumin yang merupakan antioksidan hewani, yang banyak digunakan adalah ekstrak ikan gabus (EIG) atau Channa striata. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak ikan gabus terhadap kadar F2-isoprostan pada tikus wistar model DM. Studi eksperimental ini menggunakan desain post test only control group dengan hewan coba tikus wistar jantan. Sampel yang dipilih secara acak dibagi kedalam lima kelompok perlakuan, yaitu kelompok tikus sehat (K-), kelompok tikus DM tanpa pemberian EIG (K+), kelompok tikus DM + dosis EIG 3 ml/kgBB (P1), kelompok tikus DM + dosis EIG 6 ml/kgBB (P2), kelompok tikus DM + dosis EIG 9 ml/kgBB (P3). Pemberian ekstrak dilakukan selama 8 hari. Hasil penelitian menunjukkan bahwa ada tren penurunan kadar F2-isoprostan pada kelompok P2 dan P3, meskipun pada uji Kruskal-Wallis menunjukkan signifikansi (p=0,783) yang berarti tidak ada pengaruh pemberian ekstrak ikan gabus terhadap kadar F2-isoprostan tikus model diabetes melitus.

English Abstract

Hyperglycemia that occurs in diabetes mellitus (DM) triggers the formation and increased levels of Reactive Oxygen Species (ROS) that could potentially cause oxidative stress characterized by the occurrence of lipid peroxidation. Measurement of F2-isoprostane levels can be used as a marker of oxidative stress in patients with DM. One source of antioxidants is albumin, an animal antioxidants, which is widely used is the snakehead fish (Channa striata) extract. This study aimed to determine the effect of snakehead fish extract (SFE) on F2-isoprostane levels in diabetic Wistar rat. The design of this experimental study was post-test only control group with experimental animals male Wistar rats. Samples were selected randomly divided into five treatment groups, group of healthy rats (K-), group of diabetic rats without given SFE (K +), group of DM rats + dose SFE 3 ml/kgBW (P1), group of DM rats + dose SFE 6 ml/kgBW (P2), group of DM rats + SFE dose 9 ml/kgBW (P3). The extract was given for 8 days. The results showed that there was a downward trend in the levels of F2-isoprostane groups P2 and P3, although the Kruskal-Wallis test showed significance (p = 0.783), which means there is no effect of snakehead fish extract on levels of F2-isoprostane diabetic rats.
