Abstract
Biji jintan hitam (Nigella sativa) adalah jenis tumbuhan dicotyledon yang tumbuh di negara-negara yang berbatasan dengan Laut Mediterania, Pakistan, dan India. Biji jintan hitam telah digunakan selama ribuan tahun dalam berbagai makanan seperti roti, kue-kue, yoghurt, acar, sup, saus, salad dressing, dll. Biji jintan hitam juga digunakan pada kulit atau sebagai kosmetik. Sebagian besar masalah kesehatan memiliki penyebab yang sama yaitu infeksi oleh mikro-organisme seperti bakteri, virus, parasit dan jamur. Foodborne toxicoinfections terjadi jika jasad renik yang terkonsumsi mampu menghasilkan racun sambil bereproduksi di dalam saluran cerna, salah satu contohnya adalah Shigella dysenteriae. Penyebab kontaminasi umumnya akibat hygiene sanitasi yang buruk pada penjamah makanan, selain itu juga disebabkan oleh sayur dan makanan laut. Penelitian ini bertujuan untuk membuktikan efek antimikroba ekstrak biji jintan hitam terhadap Shigella dysenteriae dengan metode dilusi agar. Metode dilusi agar digunakan untuk mengetahui nilai Kadar Hambat Minimal (KHM). Ekstrak biji jintan hitam dibuat dengan cara maserasi menggunakan etanol . Konsentrasi ekstrak biji jintan hitam yang digunakan adalah 0%, 4%, 5%, 6%, 7%, dan 8% dengan empat pengulangan. Hasil penelitian menunjukkan secara signifikan bahwa semakin tinggi konsentrasi ekstrak biji jintan hitam maka semakin rendah pertumbuhan Shigella dysenteriae (Kruskall Wallis= p<0,05; korelasi Spearman= -0,826). Kesimpulan dari penelitian ini adalah ekstrak biji jintan hitam memiliki efek antimikroba terhadap bakteri Shigella dysenteriae dengan metode dilusi agar.
English Abstract
Black cumin seeds (Nigella sativa) is a type of dicotyledon plants grown in countries bordering the Mediterranean Sea, Pakistan, and India. Black cumin seeds have been used for thousands of years in a variety of foods such as bread, cookies, yoghurt, pickles, soups, sauces, salad dressings,etc. Black cumin seeds are also used on the skin or as a cosmetic. Most of the health problems have the same cause that is infection by micro-organism such as bacteria, viruses, parasites and fungi. Foodborne toxicoinfections happen if microorganisms that consumed is able to produce toxins while reproduce in gastrointestinal tract, one of the example is Shigella dysenteriae. Cause of contamination is generally caused by poor sanitation hygiene in food handlers, but it also caused by vegetables and seafood.This experiment aims to verify the antimirobial effect of black cumin seed extract against Shigella dysenteriae with agar dilution method. Agar dilution method was used to determine the Minimal Inhibitory Concentration (MIC). Black cumin seed was extracted by maceration using ethanol. The range of Black cumin seed extract used in this research was 0%, 4%, 5%, 6%, 7%, and 8% with four repetitions. The results showed significantly that the higher concentration of black cumin seed extract, the lower the growth of Shigella dysenteriae (Kruskall Wallis = p <0.05, Spearman correlation = -0.826). The conclusion of this research is the black cumin seed extract has an antimicrobial effect against bacteria Shigella dysenteriae with agar dilution method.
