Abstract

Superoksid Dismutase (SOD) merupakan suatu antioksidan enzimatik yang melindungi sel dari stres oksidatif dengan mengkatalisa dismutase dari superokside (O2-) menjadi O2 dan H2O2. Daun ubi jalar ungu (Ipomoea Batatas L. Lam) merupakan sejenis sayuran yang mengandung antioksidan polifenol yang cukup tinggi yakni 1805 mgGAE per 100 gram. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian tepung daun ubi jalar ungu terhadap kadar SOD tikus putih (Rattus novergicus strain Wistar) yang dipapar asap rokok. Penelitian True Experimental dengan rancangan Randomized Post Test Control Only Group Design ini menggunakan sampel tikus putih yang dipelihara di Laboratorium Farmakologi FKUB. Pemilihan sampel menggunakan metode RAL (Rancangan Acak Lengkap) dengan jumlah sampel total sebanyak 30 tikus putih. Data diolah dan dianalisis menggunakan SPSS 16. Uji statistik menggunakan one way ANOVA dan uji lanjutan Post Hoc Tukey. Berdasarkan penelitian ini dapat disimpulkan bahwa terdapat penurunan kadar SOD yang signifikan antara kelompok K- dan K+ dengan p=0,000 akibat paparan asap rokok. Kadar SOD pada dosis I (0,07 gram/hari), dosis II (0,14 gram/hari), dosis III (0,28 gram/hari) tepung daun ubi jalar ungu menunjukkan peningkatan yang signifikan dibandingkan dengan kelompok K+. Hal ini membuktikan bahwa ada pengaruh pemberian tepung daun ubu jalar ungu terhadap kadar SOD pada tikus yang dipapar asap rokok. Dosis III (0,28 gram/hari) merupakan dosis efektif untuk meningkatkan kadar SOD. Dari penelitian ini disarankan adanya pengembangan pemanfaatan daun sebagai upaya diversifikasi pangan serta konsumsi daun ubi jalar ungu sebagai sumber polifenol untuk meningkatkan kadar SOD pada orang dengan kebiasaan merokok.

English Abstract

Superoxide Dismutase (SOD) is an enzymatic antioxidant that protect cells from oxidative stress by catalyzing dismutase of superoxide into O2 and H2O2. Purple sweet potato leaves (Ipomoea batatas L. Lam) is kind of vegetable plant that contain high polyphenol level, that is 1805 mgGAE on 100 gram edible portion. This research was aimed to determine the effect of purple sweet potato leaves powder to SOD level of white rat (Rattus novergicus strain Wistar) that were exposed with cigarette smoke. This True Experimental research using Post Test Control Only Group Design performed in white rat that placed in Pharmacologic Laboratory of Medical faculty of Brawijaya University. Sampling was carried out by completely random sampling with 30 rats for total sample. Data was processed and analyzed using SPSS 16. Statistical Test used one way ANOVA and Post Hoc Tukey after it. The result showed that there was a significant decrease of SOD level between K- group and K+ group (p=0,000) because of cigarette smoke exposure. The level of SOD of dosage I (0,07 gram/day), dosage II (0,14 gram/day), dosage III (0,28 gram/day) of purple sweet potato leaves powder showed a significant increasing compared to K+ group (p=0,000). It proved that purple sweet potato leaves powder has effect to white rat SOD level that is exposed with cigarette smoke. Dosage III (0,28 gram/day) was the effective dosage to increase SOD level. Based on this research, it was recommended to develop the usage of purple sweet potato as food diversification effort and consume purple sweet potato leaves as polyphenol source to increase SOD level that induced by smoking habit.
