Abstract
Avulsi adalah bentuk cedera traumatik gigi kelas lima yang didefinisikan sebagai keluarnya seluruh gigi dari soket akibat trauma. Penanganan untuk kasus avulsi adalah dengan cara melakukan replantasi secepat mungkin, jika tindakan replantasi tidak dapat dilakukan secepatnya sebaiknya gigi yang avulsi disimpan dalam suatu media penyimpanan. Media ini akan menjaga supaya sel ligamen periodontal tidak cepat mati. Penelitian ini bertujuan untuk mengatahui beda kemampuan tiga macam susu sapi UHT yang beredar di pasaran kota Malang sebagai media simpan gigi avulsi dalam menjaga viabilitas sel fibroblas secara in vitro. Jenis penelitian ini adalah eksperimental in vitro dan rancangan Randomized Post Test Only Control Group Design. Obyek perlakuan dibagi menjadi 4 kelompok yaitu, I, II, III (sebagai perlakuan) dan IV (sebagai kontrol). Sampel yang digunakan adalah kultur sel fibroblas. Sampel diinkubasi selama 24 jam kemudian media kultur diganti dengan media simpan yaitu, tiga macam susu sapi UHT yang beredar di kota Malang yaitu susu sapi UHT tanpa pewarna dan perasa, susu sapi UHT dengan pewarna dan perasa, dan susu sapi UHT tanpa merek. Jumlah sel yang hidup dalam microplate di hitung dengan ELISA reader menggunakan MTT Assay. Hasil dari penelitian didapatkan susu sapi UHT tanpa pewarna dan perasa mampu mempertahankan sel fibroblas hidup hingga 93,447%, susu sapi UHT dengan pewarna dan perasa mampu mempertahankan sel fibroblas hidup hingga 95,93% dan susu sapi UHT tanpa merek mampu mempertahankan hingga 79,243%. Kemampuan mempertahankan viabilitas ini disebabkan oleh osmolalitas dan kandungan yang terdapat pada susu sapi UHT. Dari hasil penelitian dapat disimpulkan bahwa susu sapi UHT dengan pewarna dan perasa merupakan media simpan gigi avulsi yang paling efektif dalam mempertahankan sel fibroblas.
English Abstract
Avulsion is fifth type of dental traumatic injury that is defined by tooth out of its socket becaused of trauma. Immediately replantation is the best treatment of avulsed tooth but if it cant be done, the tooth should be stored in a storage media to keep periodontal ligament cell alive. The purpose of this research is to determine effectivity differences of three variant of cow’s milk UHT which is sold in Malang’s market as storage media for avulsed tooth in preserving living fibroblast cell (in vitro). This study is experimental research (in vitro) with Randomized Post Test Only Control Group Design. Sample is divided into three group. Sample using fibroblast culture cell. First, sample is incubated for 24 hours then divided in storage media which will be tested, those are three kind of cow’s milk UHT which is sold in Malang. They are cow’s milk UHT with and without dye and flavour ingredient and unbranded cow’s milk UHT. Living Fibroblast culture cell in microplate is counted by ELISA reader using MTT Assay. The result shows cow’s milk UHT with dye and flavour ingredient can preserve living cell of fibroblast untill 95,93%, cow’s milk without dye and flavour ingredient is able to preserve living cell of fibroblast untill 93,447% and unbranded cow’s milk UHT is 79,243%.The Ability to preserve the living cell caused by their osmolalities and contents themselves. As the results, it can be concluded that cow’s milk UHT with dye and flavour ingredient is the most effective storage media for avulsed tooth in preserving living fibroblast cell.
