Abstract
Paparan asap rokok menghasilkan radikal bebas yang dapat menyebabkan stress oksidatif, sehingga dapat menginduksi pembentukan platelet yang menimbulkan risiko tromboemboli. Jumlah platelet dapat diturunkan dengan cara mengurangi stress oksidatif melalui pemberian antioksidan, salah satunya adalah katekin. Kombucha Coffee (KC) merupakan salah satu minuman yang mengandung katekin. Penelitian ini bertujuan untuk mengetahui apakah pemberian KC mampu menurunkan jumlah platelet hewan coba yang dipapar asap rokok selama 1 bulan. Penelitian ini merupakan studi eksperimental dengan menggunakan desain rancangan acak lengkap pada hewan coba tikus putih (Rattus norvegicus strain wistar). Terdapat 5 kelompok perlakuan, yaitu kontrol negatif (P0), kontrol positif (P1), paparan asap rokok + KC 2,7 mL (P2), paparan asap rokok + KC 4,05 mL (P3), paparan asap rokok + KC 5,4 mL (P4). Hasil uji statistik Oneway ANOVA menunjukkan signifikansi (Sig = 0,002 < 0,05), menandakan adanya pengaruh pemberian KC terhadap jumlah platelet tikus putih yang telah dipapar asap rokok. Dosis KC 2,7 mL dapat menurunkan platelet paling besar dibandingkan dosis lain, yaitu 21% lebih rendah dibandingkan dengan kontrol positif. Dengan demikian dapat disimpulkan bahwa pemberian KC dapat menurunkan jumlah platelet tikus putih yang telah dipapar asap rokok.
English Abstract
Cigarette smoke exposure may produce free radicals which induce oxidative stress that stimulates platelets production, and will emerge the risk of thromboembolism. Platelet count can be reduced by lowering oxidative stress through consumption of antioxidant such as catechin. Kombucha Coffee (KC) is a beverage that contains catechin. This study aims to understand the effect of KC on reducing the white rat’s platelet count exposed by cigarette smoke within a month. This research was an experimental research with completely randomized design that is done to white rats (Rattus norvegicus wistar strain) as object of experiment. There were 5 groups of intervention, those are negative control (P0), positive control (P1), cigarette smoke exposure + KC 2,7 mL (P2), cigarette smoke exposure + KC 4,05 mL (P3), cigarette smoke exposure + KC 5,4 mL (P4). Oneway ANOVA statistic analysis showed a significance (Sig = 0,002 < 0,05), which indicate that KC affect the platelet count of white rats exposed by cigarette smoke. The 2,7 mL dose of KC known to give the biggest reduction on platelet count compared to another dose, up to 21% lower than positive control. So it can be concluded that KC could reduce the platelet count of white rat exposed by cigarette smoke.
