Abstract

Diabetes melitus tipe 2 merupakan sekumpulan sindrom yang ditandai dengan kelainan metabolisme karbohidrat dan lemak karena resistensi insulin di tingkat sel. Lebih dari 75% pasien diabetes melitus tipe 2yang dirawat inap mengalami komplikasi aterosklerosis dan aterosklerosis ini menjadi penyebab kematian pada 80% pasien diabetes melitus tipe 2. Hal ini disebabkan oleh peningkatan pembentukan sel busa serta migrasi dan proliferasi sel otot polos pembuluh darah karena adanya resistensi insulin pada pasien diabetes.Kayu manis mengandung senyawa-senyawa bioaktif seperti cinnamaldehyde, methylhydroxychalcone polymer, katekin, epikatekin, prosianidin B2 serta polimer-polimer fenolik lainnyayang dapat menurunkan glukosa darah dengan cara meningkatkan pengambilan glukosa melalui translokasi GLUT4 di jaringan perifer, meningkatkan aktivitas enzim piruvat kinase dan fosfoenolpiruvat karboksikinase serta ekspresi mRNA-nyadan meningkatkan sensitivitas insulin serta menghambat pembentukan HbA1c dengan kandungan antioksidannya. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian bubuk kayu manis per oral terhadap pembentukan sel busa pada tikus putih jantan yang dibuat diabetes melitus tipe 2 dengan injeksi streptozotosin. Penelitian ini menggunakan desain true experimental randomized posttest only controlled group. Hewan coba sebanyak 25 tikus Wistar jantan dirandomisasi menjadi 5 kelompok perlakuan, yaitu: kontrol negatif, kontrol positif (model diabetes melitus tipe 2), model diabetes melitus tipe 2 dengan pemberian 3 macam dosis kayu manis (P1 27; P2 54 dan P3 108 mg/hari) selama 35 hari. Sediaan histo patologis digunakan untuk menghitung jumlah sel busa. Hasil penelitian menunjukkan bahwa tidak ada perbedaan rata-rata jumlah sel busa antarkelompok secara signifikan. Kesimpulan yang dapat diperoleh dari penelitian ini adalah pemberian bubuk kayu manis per sonde selama 40 haridapat menurunkan jumlah sel busa pada tikus model diabetes melitus tipe 2, walaupun penurunannya tidak signifikan.

English Abstract

Type 2 diabetes mellitus was a heterogeneous syndrome characterized by abnormalities in carbohydrate and fat metabolism caused by insulin resistance at the cellular level. More than 75% of all hospitalization diabetic patients had aterosclerosis and approximately 80% of all mortality in persons with diabetes mellitus was due to atherosclerosis.Insulin resistance in diabetic patients could lead to aterosclerosis by increasing foam cell formation and initiating vascular smooth muscle cell migration and proliferation.Cinnamon had several bioactive compound such as cinnamaldehyde, methylhydroxychalcone polymer, catechin, epicatechin, procyanidin B2 and phenol polymers that could lower blood glucose by increasing glucose uptake through GLUT 4 translocation in peripheral tissues, activating pyruvate kinase and phosphoenolpyruvate carboxykinase and their mRNA expression level, enhancing insulin sensitivity and inhibiting the formation of HbA1c by its antioxidants properties. The aim of this study was to examine the effect of cinnamon admission orally with foam cell formation in type 2 diabetes male Wistar rats induced by streptozotocin injection. This research used true experimental randomized posttest only controlled group design. Twenty-five male Wistar rats were randomized into five groups: control, type 2 diabetic, and type 2 diabetic with cinnamon admission in three kinds of dose (P1 27; P2 54 and P3 108 mg/day) for 35 days. The amount of foam cell were determined by histopatology preparations. The results of this research showed that the amount of foam cell in 5 groups were not different significantly. This results concluded that cinnamon admission per enteral during 40 days of study could inhibit foam cell formation in type 2 diabetes melitus rats, although the results were not significant.
