Abstract
Radikal bebas adalah molekul yang tidak memiliki pasangan elektron pada lapisan luarnya. Bentonite memiliki sisi aktif yang dapat menyerap ion fe2+ yang berperan dalam pembentukan radikal bebas. Potensi bentonite di Indonesia sebesar 380 juta ton, namun belum dimanfaatkan sebagai penyerap radikal bebas yang diakibatkan herbisida paraquat. Tujuan penelitian ini untuk mengetahui pengaruh bentonite terhadap penurunan kadar malondialdehyde (MDA) jaringan otak Rattus novergicus yang dipapar herbisida paraquat. Penelitian ini menggunakan true experimental design dengan metode yang digunakan yaitu randomized posttest only controlled group design. Dua puluh tikus dibagi menjadi lima kelompok tikus, yaitu kelompok tidak dipapar paraquat (P0), kelompok yang dipapar paraquat (P1), kelompok yang dipapar paraquat dan bentonite 10% (P2), kelompok yang dipapar paraquat dan bentonite 20% (P3), dan kelompok yang dipapar paraquat dan bentonite 30% (P4). Paraquat dan bentonite diberikan selama 8 hari secara per oral. Kemudian kadar MDA otak diukur metode thiobarbituric acid (TBA). Analisa data dengan metode ANOVA terdapat perbedaan bermakna (P=0,000) rerata kadar MDA antar kelompok. Selanjutnya dilakukan uji post hoc Tukey terdapat perbedaan bermakna (p<0,05) pada semua kelompok kecuali kelompok P0 dan P3, P0 dan P4, P3 dan P4. Hasil penelitian menunjukkan adanya pengaruh bentonite terhadap penurunan kadar MDA jaringan otak Rattus Norvegicus yang dipapar herbisida paraquat.
English Abstract
ree radicals are molecules that do not have a pair of electrons in its outer layers. Bentonite has an active side which can absorb Fe2+ ions that play a role in the formation of free radicals. Potency of bentonite in Indonesia is 380 million ton but did not use to absorb free radical from herbicide paraquat. This study aimed to determine the effect of bentonite MDA rate of brain’s tissue rattus novergicus that exposed by herbicide paraquat. This study used true experimental design with Randomized Posttest Only Controlled Group Design. Twenty rats divide to five groups of rats, the group not exposed to paraquat (P0), the group exposed to paraquat (P1), the group exposed to paraquat and bentonite 10% (P2), the group exposed to paraquat and bentonite 20% (P3), and group exposed to paraquat and bentonite 30% (P4). Paraquat and bentonite are given for 8 days perorally). Then the MDA rate od brain’s tissue were measured with thiobarbituric acid method (TBA). The data was analyzed with ANOVA there was a significant difference (P=0,000) in MDA rate of brain’s tissue between groups. And then analyzed by Tukey post hoc test show a significant difference (p<0.05) in all groups except the group P0 and P3, P0 and P4, P3 and P4. From these results we can conclude that the effect of bentonite is decrease MDA rate of brain’s rattus novergiicus tissue that exposed by herbicide paraqua
