Abstract
Diabetes mellitus (DM) tipe 2 merupakan penyakit metabolik dengan karakteristik kadar gula darah meningkat yang terjadi karena penurunan progresif sekresi insulin oleh sel beta pankreas dan terjadinya resistensi insulin. Pada progresifitas penyakit DM tipe 2 terjadi peningkatan kemokin proinflamasi pada pankreas maupun sel perifer yang akan menyebabkan inflamasi. Inflamasi ini akan mengurangi kemampuan sel beta pankreas untuk beregenerasi dan menyebabkan penurunan produksi insulin serta terjadinya resistensi insulin. Nikotin diduga memiliki efek anti-inflamasi dan mudah untuk diperoleh di Indonesia (diambil dari daun tembakau (Nicotiana tabacum)). Penelitian bertujuan untuk mengetahui tentang pengaruh pemberian ekstrak etanol daun Nicotiana tabacum terhadap peningkatan produksi insulin berdasarkan penghitungan HOMA Beta pada model tikus Rattus norvegicus galur Wistar yang diinduksi diabetes mellitus tipe 2 dengan High Fat Diet selama 4 minggu dan STZ injeksi. Sampel dibagi menjadi 5 kelompok, “normal”, “DM tipe II”, “DM tipe II dengan ekstrak etanol daun tembakau 90mg/kgBB”, “DM tipe II dengan ekstrak 180mg/kgBB”, dan “DM tipe II dengan ekstrak 270mg/kgBB”. Pemberian ekstrak dilakukan selama 4 minggu secara oral. Setelah itu diambil darahnya untuk dilakukan pengukuran HOMA Beta. Hasil penelitian menunjukkan peningkatan perhitungan HOMA Beta secara signifikan dengan dosis 180mg/kgBB dapat meningkatan produksi insulin hampir setara dengan tikus normal.
English Abstract
Diabetes mellitus (DM) type 2 is a metabolic disease characterized by elevated blood sugar levels due to a progressive decrease in insulin secretion by pancreatic beta cells and increase of insulin resistance. In the progression of type 2 diabetes mellitus there is an increase in proinflammatory chemokine in the pancreas and peripheral cells that cause inflammation. This inflammation will reduce the ability of pancreatic beta cells to regenerate and cause decreased insulin production as well as the occurrence of insulin resistance. Nicotine is thought to have an anti-inflammatory effect and is easy to obtain in Indonesia (taken from the leaves of tobacco (Nicotiana tabacum)). This research aims to know about the effect of etanol extract of Nicotiana tabacum leaves on the increase of insulin production based on HOMA Beta calculation on rats model Rattus norvegicus Wistar strain induced type 2 diabetes mellitus with high fat diet for 4 weeks and injection of STZ. Samples were divided into 5 groups, "normal", "Type II DM", " Type II DM with etanol extract of Nicotiana tabacum leaves 90mg/kgBB ", " Type II DM with 180mg/kgBB extract ", and " Type II DM with 270mg/kgBB extract " groups. Extract given for four weekks orally. After that, rats blood was taken to measure HOMA Beta. The results showed a significant increase in HOMA Beta calculation with a dose of 180mg / kgBB could increase the production of insulin almost equivalent to normal rats.
