Abstract
Xilanase merupakan enzim yang mengkatalisis reaksi hidrolisis xilan menjadi xilosa. Xilanase bebas tidak dapat digunakan lebih dari satu kali sehingga perlu dilakukan amobilisasi. Xilanase diamobilisasi menggunakan metode adsorpsi dengan zeolit dan penjebakan dengan Ca-alginat. Penelitian ini bertujuan untuk mengetahui konsentrasi xilanase optimum dan konsentrasi alginat optimum dengan cara menentukan kadar protein dan aktivitas enzim optimum dengan melakukan variasi konsentrasi xilanase dan konsentrasi alginat serta menentukan efisiensi pemakaian ulang xilanase amobil. Pada penelitian ini dilakukan variasi konsentrasi xilanase (2; 2,5; 3; 3,5; 4) mg/mL dan variasi konsentrasi alginat (1,5; 2; 2,5; 3; 3,5) % w/v. Kadar protein diukur dengan menggunakan metode spektrofometri menggunakan reagen biuret dan aktivitas gula pereduksi dengan menggunakan reagen DNS. Kondisi optimum amobilisasi xilanase pada konsentrasi xilanase 3,5 mg/mL dengan jumlah enzim yang terjebak 16,68 mg dan aktivitas enzim sebesar 28,02 μg mg-1 menit -1 dan konsentrasi alginat 3 % w/v dengan jumlah enzim yang terjebak 15,91 mg dan aktivitas sebesar 11,33 μg mg-1 menit
English Abstract
Xylanase is an enzyme that catalyzes the reaction of xylan hydrolysis into xylose products. Free xylanase can not be used more than once in an application, so that need to immobilization. Xylanase was immobillized using adsorption with zeolit and trapping with Zeolite Ca-alginat-chitosan method. This research aim to determine optimum alginate concentration and optimum xylanase concentration by determining protein level and optimum xylanase activity in vary concentration of alginate and concentration of xylanase, and also to determine reuse eficiency of immobilized xylanase. This research used variation of xylanase (2; 2,5; 3; 3,5; 4) mg/mL and the alginate concentration (1,5; 2; 2,5; 3; 3,5) % w/v. Protein level was determined with spectrophotometry method by using biuret reagent and activity of reducing sugar was determined by using DNS reagent. Optimum condition of immobilized xylanase was on xylanase concentration is 3,5 mg/mL with trapped enzyme was 16,68 mg and activity was 28,02 μg mg-1 menit -1 and 3% w/v alginate concentration with trapped enzyme was 15,91 mg and activity was 11,33 μg mg-1 menit -1.
