Pengaruh Ekstrak Kulit Tomat Terhadap Degenerasi Hepatosit Pada Tikus Model Diabetes Mellitus Tipe 2

Abstract

Jumlah penderita DM tipe 2 terus meningkat di seluruh dunia. DM tipe 2 menimbulkan banyak komplikasi, salah satunya adalah degenerasi hepatosit karena terdapat peningkatan radikal bebas yang menyebabkan stress oksidatif. Kulit tomat mempunyai kandungan antioksidan yang dapat membersihkan radikal bebas. Penelitian ini bertujuan untuk mengetahui adanya pengaruh pemberian ekstrak kulit tomat terhadap degenerasi hepatosit pada tikus (Rattus norvegicus) galur Wistar jantan model DM tipe 2. Desain penelitian ini menggunakan Post Test Randomized Control. Tikus Wistar jantan dibagi menjadi 5 kelompok, yaitu KN(diet normal), KP (STZ dan diet tinggi lemak), KP1 (STZ,diet tinggi lemak, dan ekstrak kulit tomat 50mg/kgBB), KP2 (STZ,diet tinggi lemak, dan ekstrak kulit tomat 100mg/kgBB), dan KP3 (STZ,diet tinggi lemak, dan ekstrak kulit tomat 150mg/kgBB). Tikus kemudian dibedah dan diambil heparnya untuk dibuat preparat histologi. Perubahan yang dilihat adalah degenerasi hepatosit (dengan ciri-ciri sel bervakuola, sel membengkak, dan atau sitoplasma sel yang mengeruh) per lima lapang pandang yang lalu jumlahnya dibandingkan dengan jumlah keseluruhan sel dan dikali 100% untuk mendapatkan angka persentasenya. Data lalu dianalisa dengan uji statistik One-way Anova dilanjutkan dengan Post-hoc. Hasil penelitian menunjukkan bahwa ada peningkatan persentase degenerasi hepatosit yang signifikan pada KP dibandingkan dengan KN. Serta adanya penurunan persentase degenerasi hepatosit yang signifikan pada KP3 dibandingkan dengan KP. Maka dapat disimpulkan bahwa ekstrak kulit tomat dosis 150mg/kgBB dapat mencegah degenerasi dari hepatosit pada DM2.

English Abstract

The number of people with type 2 diabetes continues to increase worldwide. Type 2 diabetes presents many complications, one of which is hepatocyte degeneration because there is an increase of free radicals that cause oxidative stress. Tomato skin contains antioxidants that can clean up the free radicals. This study aims to determine the effect of tomato skin extract on hepatocyte degeneration in rat (Rattus norvegicus) male Wistar strain model DM type 2. The design of this study using Post Test Randomized Control. Male Wistar rats were divided into 5 groups, namely KN (normal diet), KP (STZ and high-fat diet), KP1 (STZ, high-fat diet, and tomato skin extract 50mg / kgBB), KP2 (STZ, high fat diet, and 100mg / kgBB tomato skin extract), and KP3 (STZ, high fat diet, and 150mg / kgBB tomato skin extract). The rats were then dissected and taken up for making preparations of histology. The changes seen are hepatocyte degeneration (with the characteristics of vacuolated cells, swollen cells, and / or cloudy cell cytoplasm) per fifth of the viewing area then the number compared with the total number of cells and multiplied by 100% to obtain the percentage. The data is then analyzed by One-Way Anova statistic test followed by Post-hoc. The results showed that there was a significant increase of percentage of hepatocyte degeneration in KP compared with KN. As well as a significant decrease in the percentage of hepatocyte degeneration in KP3 compared with KP. So it can be concluded that tomato skin extract dose 150mg / kgBB can prevent degeneration of hepatocytes in DM2.
