Abstract
Kanker serviks adalah penyakit kanker dengan tingkat kematian terbanyak kedua di dunia dan penyebab kematian tertinggi akibat kanker di Indonesia. Penelitian ini bertujuan untuk melakukan sintesis ester eugenol dengan starting material eugenol, melaksanakan uji antiproliferatif dan mengukur kemampuan ester eugenol dalam apoptosis sel kanker serviks. Isolasi eugenol dari minyak cengkeh dilakukan dengan dua cara yaitu reaksi asam basa dan distillasi fraksinasi dengan kemurnian hasil eugenol dari distillasi fraksinasi 100% dan rendemen 70,66%. Sintesis ester eugenol dilakukan dengan prinsip reaksi adisi-esterifikasi melalui reaksi antara eugenol hasil isolasi dengan asam format dengan perbandingan mol 1:27 dan lama reaksi 11 jam. Analisis data GC-MS terhadap hasil sintesis menunjukkan ester eugenol dihasilkan dengan kemurnian 92,42% dengan persen yield sebesar 93,34%. Data spektra UV dari ester eugenol menunjukkan pembentukan gugus karbonil (gugus C≡O) dan hilangnya serapan yang menunjukkan hilangnya ikatan rangkap gugus vinil dalam struktur eugenol. Dari hasil pengamatan cytogram ditentukan nilai LC50 dari ester eugenol berada pada konsentrasi 48,73 ppm yang menunjukkan bahwa kemampuan apoptosis yang sangat kuat dimana dengan konsentrasi di bawah 50 ppm ester eugenol mampu menginduksi apoptosis sel HeLa kanker serviks. Oleh karena itu dapat disimpulkan bahwa gugus karbonil yang ada di ester eugenol aktif sebagai antioksidan dan berpotensi sebagai antikanker.
English Abstract
Cervical cancer is a cancer with the second most death rate in the world and the highest cause of death from cancer in Indonesia. This study is aimed to perform eugenol ester synthesis with eugenol starting material, carry out antiproliferative test and measure the ability of eugenol ester in apoptosis of cervical cancer cells. Isolation of eugenol from clove oil was done in two ways: acid-base reaction and fractionation distillation with eugenol purity of 100% fractionation distillation and 70.66% yield. The eugenol ester synthesis was carried out with the principle of esterification-addition reactions through the reaction between the isolated eugenol and the formic acid with the mole ratio of 1:27 and the reaction time of 11 hours. Analysis of GC-MS data on the synthesis results showed that eugenol ester was produced with a purity of 92.42% with a yield percentage of 93.34%. UV spectra data from the eugenol esters showed the formation of carbonyl groups (C≡O groups) and loss of absorption showing the loss of double bonds of vinyl groups in the eugenol structure. From the observation of the cytogram determined the value of LC50 from the eugenol ester was at 48.73 ppm concentration which showed that very strong apoptosis ability where the concentration belowed 50 ppm of ester eugenol was able to induce hela cell cervical cancer apoptosis. The conclusion is the carbonyl group present in the eugenol ester is active as an antioxidant and potentially as an anticancer.
