Abstract
Peptic ulcer disease (PUD) merupakan kondisi yang muncul saat lapisan mukosa protektif dari mukosa gastrointestinal mengalami kerusakan. Lapisan protektif pada mukosa lambung berupa mucus dan bikarbonat dapat rusak akibat asam lambung dan pepsin yang berasal dari usus dua belas jari. Kerusakan lapisan protektif juga dapat diakibatkan oleh penggunaan NSAID melalui mekanisme defisiensi prostaglandin. Hingga saat ini, penghentian NSAID merupakan langkah pertama pencegahan PUD. Sargassum sp., salah satu jenis rumput laut yang terbukti memiliki efek proteksi terhadap interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-10 (IL-10), menormalkan aktivtas enzim antioksidan, serta menghambat lipid peroxidation pada mukosa lambung. Sargassum mengandung agen yang memiliki efek anti ulkus, dengan cara menjaga volume serta keasaman dari cairan lambung, dan meningkatkan sistem pertahanan antioksidan mukosa lambung pada model hewan coba gastritis menggunakan ethanol. Selain itu, efek anti ulkus yang dimiliki oleh Sargassum dicapai melalui pembentukan prostaglandin endogen. Penelitian ini adalah true experimental dengan metode Randomized Post Only Controlled Group Design, yang dilakukan secara in vivo menggunakan tikus yang diinduksi indometasin. Dari penelitian yang telah dilakukan menunjukkan bahwa pemberian ekstrak Sargassum sp. mampu menurukan skor ulkus lambung secara signifikan. Terdapat penurunan skor ulkus lambung yang signifikan antara kontrol positif dengan ekstrak Sargassum sp. dosis 150 mg/kgBB, 300 mg/kgBB, 600 mg/kgBB, maupun misoprostol 90 mcg/kgBB. Dosis optimum pencegahan ulkus lambung adalah ekstrak alga cokelat 150 mg/kgBB.
English Abstract
Peptic ulcer disease (PUD) is a condition that occurs when the protective mucosal layer of the gastrointestinal mucosa is damaged. The protective layer of the gastric mucosa of mucus and bicarbonate can be damaged by gastric acid and pepsin originating from the duodenum. Protective layer damage can also be caused by the use of NSAID through prostaglandin deficiency mechanisms. To this end, the termination of NSAID is the first step of PUD prevention. Sargassum sp., A type of seaweed that is shown to have protective effects on interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-10 (IL-10), normalize antioxidant enzyme activity, and inhibit lipid peroxidation of the gastric mucosa. Sargassum contains agents that have anti-ulcer effects, keeping the volume and acidity of the stomach fluid, and improving the gastric mucosal antioxidant defense system in animal models of gastritis using ethanol. In addition, the anti-ulcer effects possessed by Sargassum are achieved through the formation of endogenous prostaglandins. This study was true experimental with Randomized Post Only Controlled Group Design method, which was conducted in vivo using indomethacin-induced mice. From the research that has been done shows that giving Sargassum sp. able to significantly decrease stomach ulcer score. There was a significant decrease in gastric ulcer score between positive control with Sargassum sp. extract dose 150 mg/kgBB, 300 mg/kgBB, 600 mg/kgBB, or misoprostol 90 mcg/kgBB. The optimum dosage of gastric ulcer prevention is chocolate algae extract 150 mg/kgBB.
