Abstract

Tujuan dari penelitian ini adalah untuk mengetahui pengaruh escalating dose self-antigen immunotherapy menggunakan self-antigen dsDNA terhadap regulasi sistem imun dengan mengukur kadar ANA pada mencit lupus induksi pristane. 20 mencit Balb/C betina berusia 6-8 minggu dibagi menjadi 2 kelompok: 16 mencit mendapat injeksi tunggal pristane intraperitoneal (i.p.) 0,5 cc untuk induksi lupus dan 4 mencit sebagai kontrol. Dimulai pada 12 minggu setelah injeksi, 12 mencit yang diinduksi oleh pristane dibagi menjadi tiga kelompok berdasarkan dosis dsDNA yang diberikan secara i.p. selama tiga minggu: Kelompok A (0,005 μg, 0,05 μg, 0,5 μg), kelompok B (0,05 μg, 0,5 μg, 5 μg) dan kelompok C (0,5 μg, 5 μg, 50 μg). Dosisnya ditingkatkan sepuluh kali tiap minggu. dsDNA dikomplekskan dengan polietilenimin polimer kationik (PEI) 2 sebelum diinjeksikan. Sebanyak 20 mencit Balb/C dianalisis untuk serum kadar antinuclear antibody (ANA) menggunakan metode enzyme-linked immunosorbent assay (ELISA). Kadar ANA pada kelompok yang diberikan injeksi dsDNA mengalami penurunan tetapi tidak ada perbedaan yang signifikan dengan kelompok kontrol negatif. Kesimpulannya, meskipun hipotesis tidak terbukti secara statistik, masih terdapat kecenderungan penurunan pada kadar ANA. Selain itu, beberapa kelemahan pada penelitian ini juga perlu diperbaik seperti lama durasi induksi lupus serta terapi yang terlalu singkat menyebabkan daya desensitisasi self-antigen dsDNA yang tidak optimal, dan pemilihan tehnik sampling yang kurang tepat.

English Abstract

The aim of this study was to develop a novel therapeutic method of escalating dose self-antigen immunotherapy using self-antigen dsDNA against immune system regulation by measuring ANA levels in pristane-induced lupus mice model. 20 female Balb/C mice, 6–8 weeks old were divided into 2 groups: 16 mice received a single intraperitoneal (i.p.). injection of 0.5 cc pristane for lupus induction and 4 mice as healthy controls. Starting at 12 weeks after injection, 12 pristane-induced lupus mice were divided into three groups based on doses of dsDNA received intraperitoneally for three weeks: Group A (0,005 μg, 0,05 μg, 0,5 μg), group B (0,05 μg, 0,5 μg, 5 μg) and group C (0,5 μg, 5 μg, 50 μg). The doses would be increased ten times every week. dsDNA were complexed with the cationic polymer polyethylenimine (PEI)2 before injection. A total of 20 Balb/C mice were analyzed for level of antinuclear antibody (ANA) in serum using enzyme-linked immunosorbent assay (ELISA). Level of ANA in the serum decreased unsignificantly. In conclusion, although the hypothesis is not proven statisitically, however level of ANA still shows a decreased trend in this study. Besides, there are some weaknesses that are need to be fixed in this study such as the duration of lupus induction and the treatments given that was too short causing desensitization with self-antigen dsDNA is not optimal, and the imprecise sampling technique used
