Abstract
Diabetes melitus (DM) tipe 2 merupakan penyakit metabolik kronis yang ditandai oleh terjadinya resistensi insulin dan menurunnya fungsi sel β pankreas. Treadmill merupakan salah satu terapi yang dapat diberikan pada penderita DM tipe 2 untuk menurunkan kadar glukosa darah. Penelitian ini bertujuan untuk mengetahui pengaruh treadmill terhadap kadar malondialdehyde (MDA) dan gambaran histopatologi hepar pada tikus (Rattus norvegicus) model diabetes melitus tipe 2. Dalam penelitian ini, resistensi insulin terjadi akibat pemberian HFD selama sembilan minggu dan injeksi streptozotocin (STZ) untuk menginduksi terjadinya hiperglikemia yang merupakan salah satu gejala penyakit DM tipe 2. Penelitian eksperimental ini menggunakan 18 ekor tikus Wistar jantan dengan berat badan 100-150 gram yang dibagi menjadi tiga kelompok, yaitu 6 ekor kelompok kontrol negatif (K-), 6 ekor kelompok kontrol positif (K+) dengan induksi HFD dan STZ tanpa perlakuan treadmill, dan 6 ekor kelompok perlakuan (P) dengan induksi HFD dan STZ serta diberikan perlakuan treadmill. Kadar MDA diuji dengan menggunakan metode tes Thiobarbituric Acid Reactive Substances (TBARS test) kemudian dianalisis secara kuantitatif menggunakan uji one way ANOVA. Sedangkan gambaran histopatologi hepar diwarnai dengan pewarnaan Hematoxylin Eosin (HE) kemudian dianalisa secara kualitatif deskriptif. Hasil penelitian menunjukkan bahwa treadmill tidak berpengaruh secara signifikan terhadap penurunan kadar MDA hepar, namun terjadi tren penurunan rata-rata kadar MDA hepar pada tikus DM treadmill sebesar 8,15% terhadap tikus DM (kontrol positif). Sedangkan terhadap gambaran histopatologi hepar, treadmill dapat mengurangi akumulasi fatty liver tikus DM treadmill terhadap tikus DM (kontrol positif).
English Abstract
Type 2 Diabetes melitus is a chronic metabolic disease caused by insulin resistance and decreasing in pancreatic β cell function. Treadmill was a one of therapy which can be given to type 2 DM patients to lowering the levels of blood glucose. This research purposed to know the effect of treadmill against the level of malondialdehyde (MDA) and liver histopathology in rat (Rattus norvegicus) with type 2 diabetes melitus model. In this research, insulin resintance occur due to giving HFD for nine weeks and streptozotocin (STZ) injection to induced the happening of hiperglikemia which is one of the symptoms of Type 2 DM disease. This experimental research is using 18 male wistar rats with a weight of 100-150 grams which divided into three groups, which is six rats is negative control group (K-), another six rats is postive control group with HFD and STZ induced without treadmill treatment (K+), and the last six rats is HFD and STZ induced with treadmill treatment (T). The levels of MDA tested with using Thiobarbituric Acid Reactive Substances (TBARS) Test and then analyzed in quantity using one way ANOVA test. Meanwhile liver histopathology coloured with Hematoxylin Eosin (HE) and then analyzed in qualitative and descriptive. The result of the research showed that treadmill has no significant effect against decreasing of amount of liver MDA, however decreasing of liver MDA average of DM treadmill Rats about 8,15% impact to DM rats (positive control). Meanwhile in liver histopathology, treadmill can reduce the accumulation of fatty liver in DM treadmill rats impact to DM rats (positive control).
