Abstract

Hati merupakan organ penting tubuh manusia karena berperan pada sekresi empedu dan pengolahan metabolik nutrien utama. Sel-sel hati meregenerasi secara cepat dapat mempertahankan fungsinya bila terjadi gangguan ringan, namun dapat berakibat fatal apabila terjadi gangguan berat. Kerusakan hati masih menjadi salah satu penyakit penyebab kematian di Indonesia. Salah satu penyebabnya akibat stres oksidatif, yaitu ketidakseimbangan radikal bebas dengan antioksidan yang menyebabkan kerusakan sel akibat konsumsi alkohol maupun obat-obatan yang berlebih. Untuk mengatasi hal tersebut, maka dipilih serbuk ekstrak air sarang semut (Myrmecodia tuberosa) sebagai hepatoprotektor yang mengandung senyawa bioaktif dan memiliki aktivitas antioksidan. Tujuan penelitian ini adalah mengetahui pengaruh konsumsi dan pemberian dosis terbaik serbuk ekstrak air sarang semut pada tikus wistar yang diinduksi etanol terhadap kadar enzim SGOT, SGPT, SOD dan MDA serta kondisi histopatologi hati. Penelitian ini dilakukan melalui dua tahap. Tahap pertama yaitu pembuatan serbuk ekstrak air sarang semut. Tahap kedua yakni pengujian serbuk ekstrak air sarang semut sebagai hepatoprotektor secara in vivo terhadap kadar enzim SGOT, SGPT, SOD dan MDA serta histopatologi hati menggunakan Rancangan Acak Lengkap (RAL). Data yang diperoleh di analisa dengan menggunakan Analysis of Variant (ANOVA). Selanjutnya dilakukan uji lanjut BNT. Hasil penelitian menunjukkan bahwa serbuk ekstrak air sarang semut memiliki rendemen sebesar 23,37%, kadar air sebesar 4,67%, kandungan fenol sebesar 5,324 mg/g, total tanin sebesar 3,727 mg/g, total flavonoid 10,336 mg/g dan aktivitas antioksidan IC50 sebesar 187,34 ppm. Hasil penelitian secara in vivo menunjukkan pemberian serbuk ekstrak air sarang semut memberikan pengaruh nyata (α=0,05) terhadap penurunan kadar SGOT, SGPT, MDA dan peningkatan SOD serta perbaikan gambaran histopatologi l hati. Hasil menunjukkan bahwa peningkatan paling rendah setelah pemberian larutan etanol diperoleh pada dosis III (1890 mg/kg BB), yaitu dengan kenaikan kadar SGOT sebesar 9,784%, kadar SGPT 0,946%, kadar SOD sebesar 6,228 U/ml dan kadar MDA sebesar 805 ng/ml serta perbaikan sel hati yang ditunjukkan dengan adanya tipe open face nucleus pada sel hati.

English Abstract

Liver is an important organ of the human body which plays roles in bile secretion and nutrient metabolism. The liver cells, which has the ability to regenerate rapidly, can maintain their function in the event of mild disturbance but may be fatal to the human body if severe disturbance happens. Liver failure is still one of the leading causes of death in Indonesia. One of the causes is oxidative stress, which is an imbalance of the number of free radicals with the amount of antioxidants that may damage the cell due to excessive consumption of alcohol and drugs. To outgrow the problem, white ant nest (Myrmecodia tuberosa) water extract powder is selected as one of hepatoprotector agent containing bioactive compound, and also its possesion of antioxidant activity. This study aims to determine the effect of consumption and the best dosage of white ant nest water extract powder given to ethanol-induced wistar rats towards the level of SGOT, SGPT, SOD enzymes, MDA, and histopathologic conditions of the liver. This research is done through two stages. The first stage is the ant nest water extract powder manufacture, and the second stage is the in vivo examination of ant nest water extract powder as hepatoprotector towards, the level of SGOT, SGPT, SOD, MDA, and histopathologic conditions of the liver using One-factorial Randomized Complete Design (RCD). The obtained data is then analyzed using Analysis of Variant (ANOVA). If the result shows significant difference, the data is further analyzed using LSD (Least Significat Difference) Test. The result showed that the white ant nest yielded out 23,37% of water extract with 4,67% of water content. The bioactive profile of the extract showed 5,324 mg/g of phenol content, 3,727 mg/g of total tannin, 10,336 mg/g of flavonoid, and 187,34 ppm of IC50 antioxidant activity. The in vivo examination showed that the ant nest water extract powder gave significant effect (α=0,05) to the decreased levels of SGOT, SGPT, MDA, also the improvement of SOD and hispathologic features of liver cells. The in vivo test showed the lowest weight increase after ethanol was induced falls to dose III (1890 mg/kg body weight), which increased 9,784% of SGOT level, 0,946% SGPT level, 6,228 U/ml of SOD enzymes and 805 ng/ml of MDA. The improvement of liver cells is shown by the existence of open face type of nucleus in liver cells.
