Abstract

Sardinella lemuru merupakan ikan tangkapan pelagis penting tidak hanya di Selat Bali, akan tetapi juga di Selatan (Grajagan dan Puger) dan Utara (Selat Madura) Jawa Timur. Hasil tangkapan lemuru di Perairan Probolinggo dan Puger selama 5 tahun terakhir (2010-2014) mengalami penurunan diduga karena Sardinella lemuru bermigrasi ke wilayah perairan lain di Samudera Hindia. Kondisi lingkungan migrasi yang berbeda dapat mempengaruhi kemampuan adaptasi Sardinella lemuru dan perpindahan gen antar populasi. Keragaman genetik yang rendah akan menurunkan kemampuan adaptasi suatu populasi. Analisis genetik dengan teknik sekuensing dipilih dikarenakan lebih efisien, terus berkembang dan dapat dipercaya. Analisis keragaman genetik pada daerah mitokondria control region (non coding) dipilih karena lebih hipervariabel dibandingkan daerah coding dan sebelumnya belum pernah dilakukan penelitian genetik Sardinella lemuru pada daerah control region di Perairan Probolinggo dan Puger, sehingga diperlukan penelitian untuk mengetahui keragaman, kekerabatan genetik dan stok populasi. Sardinella lemuru dari fishing ground Probolinggo dan Puger (total 40 ekor ikan) diambil sampel jaringan, ekstraksi DNA, PCR (primer CRK dan CRE), elektroforesis dan sekuensing. Hasil analisis keragaman haplotipe Sardinella lemuru Probolinggo dan Puger tinggi (Hd=1) dan hubungan kekerabatan antara populasi jauh (jarak genetik= 0,321). Kekerabatan secara genetik dan morfologi Sardinella lemuru membentuk dua Clade yang mewakili subpopulasi Utara dan Selatan Jawa dengan tingkat aliran gen antara populasi rendah (Nm= 0,45). Populasi Sardinella lemuru Probolinggo, 6 spesies Puger dan out group mewakili karakter dominan (nenek moyang) dan 14 spesies Puger mewakili karakter alternatif (keturunan). Tingkat kedekatan antar populasi dipengaruhi oleh spesies kriptik dan pergerakan arus musiman di Perairan Probolinggo dan Puger.

English Abstract

Sardinella lemuru pelagic fish catch is important not only on the strait of Bali , but also in Southern (Grajagan and Puger) and the Northern (the strait of Madura) in East Java . The catch in the Probolinggo and lemuru Puger during 5 years (2010-2014) decline might have been caused by Sardinella lemuru migrate to other territorial waters in the indian ocean. Environmental conditions migration different can affect the adaptation Sardinella lemuru and displacement genes between population. Low genetic diversity would decrease the ability of adaptation of a population. Genetic analysis to technique sequencing chosen is because more efficient , continue to grow and reliable. Analysis in the mitochondria genetic diversity to the areas which control region (non coding) were selected because it is hipervariabel coding and formerly from the genetic research has never been undertaken Sardinella lemuru to the areas which control a region in Probolinggo and Puger waters , research to know the diversity that they need, genetic kinship and stock the population Sardinella lemuru of fishing ground Probolinggo and Puger (total 40 the tail of a fish) taken a tissue sample , the extraction of DNA, PCR (CRK and CRE) primary, and sekuensing electrophoresis. The analysis diversity haplotipe Sardinella lemuru Probolinggo and Puger high (Hd = 1) and relationship kinship the population far (genetic distance = 0,321). The genetically and morphology Sardinella lemuru form two clade representing sub-population North and South Java with levels of genes the population low (Nm = 0,45). Population Sardinella lemuru Probolinggo, 6 species Puger and out group representing the character dominant (ancestors) and 14 species Puger represent characters alternate (ancestry). The level of proximity between the population affected by a species of cryptic and movement of currents seasonal Probolinggo and Puger waters.
