Abstract
Diazinon merupakan insektisida jenis organofosfat yang banyak digunakan pada sebagian besar sektor pertanian. Hasil metabolisme senyawa organofosfat ini dapat meningkatkan Reactive Oxygen Species (ROS) dalam tubuh, sehingga akan menyebabkan komplikasi ke berbagai organ, termasuk lambung. Ekstrak kulit buah naga merah (Hylocereus polyrhizus) memiliki aktivitas antioksidan yang tinggi dalam menurunkan ROS pada kasus keracunan diazinon. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak kulit buah naga merah dalam menurunkan kadar MDA dan memperbaiki kerusakan gambaran histopatologi lambung tikus yang diinduksi diazinon. Penelitian ini menggunakan tikus putih (Rattus norvegicus), jantan strain Wistar berumur 8-12 minggu, berat badan rata-rata 150 g sebagai hewan coba sebanyak 20 ekor. Terdapat 5 kelompok perlakuan, yaitu kelompok K(-), K(+), P1, P2, dan P3. Dosis diazinon yang digunakan sebanyak 40 mg/kg BB per-oral selama 5 hari berturut-turut. Ekstrak kulit buah naga merah (Hylocereus polyrhizus) yang diberikan dengan dosis bertingkat yaitu 150 mg/150 g BB, 200 mg/150 g BB, dan 250 mg/150 g BB per-oral, selama 14 hari. Data kadar MDA dianalisis kuantitatif dengan ANOVA menggunakan program komputer SPSS version 22 for Windows serta dilanjutkan dengan uji Beda Nyata Jujur (BNJ) α = 5% dan gambaran histopatologi lambung dianalisis secara deskriptif. Hasil penelitian menunjukkan pemberian ekstrak kulit buah naga merah (Hylocereus polyrhizus) dosis 150 mg/150g BB secara sangat signifikan (p<0.01) sudah mampu menurunkan kadar MDA lambung dan mampu memperbaiki gambaran histopatologi labung.
English Abstract
Diazinon is an organophosphate type insecticide widely used in most agricultural sectors. The metabolic results of these organophosphate compounds can increase Reactive Oxygen Species (ROS) in the body, thus causing complications to the various organs of one of the stomach. Red pitaya peel extract has high antioxidant activity in lowering ROS, in cases of diazinon poisoning. This study aims to determine the effectiveness of Red-pitaya peel extract in reducing levels of Malondialdehyde (MDA) and repair histopathological damage to rat induced diazinone. This study used white rats (Rattus norvegicus) male Wistar strain aged 8-12weeks, average weight 150 g as a trial animal of 20 tails. There were 5 treatment groups, which were K (-), K (+), P1, P2, and P3. The dose of diazinone used was 40 mg / kg BW with oral sonde for 5 consecutive days. Red pitaya peel (Hylocereus polyrhizus) extract given with a multilevel dose of 150 mg/150 g BW, 200/150 g BW mg and 250 mg/150 g BW, for 14 days. MDA levels were measured using the Thiobarbituric acid (TBA) test and gastric histopathology features using Hematoxylin-Eosin (HE) staining. The MDA content was analyzed quantitatively by ANOVA using SPSS version 22 computer program for Windows and continued with honestly significant difference (HSD) test (α = 5%) and gastric histopathology were analyzed descriptively. The results showed that the extract of red pitaya peel (Hylocereus polyrhizus) dose 150 mg/150 g BW significantly (p<0.01) has been able to reduce levels of MDA gastric dan improve the histopthology of the stomach.
