Abstract

Karies gigi merupakan proses demineralisasi yang menyerang jaringan keras gigi disebabkan oleh aktivitas mikroba yang meragikan karbohidrat. Lactobacillus acidophilus merupakan salah satu bakteri yang berperan penting dalam karies gigi. Daun pandan merupakan salah satu bahan alami yang dapat berfungsi sebagai antibakteri karena mengandung bahan aktif seperti flavonoid, alkaloid, tannin, dan saponin. Penelitian ini bertujuan untuk membuktikan terdapat pengaruh antibakteri daun pandan terhadap bakteri Lactobacillus acidophilus secara in vitro. Penelitian ini merupakan penelitian eksperimental laboratorik dengan menggunakan metode dilusi tabung untuk menentukan Kadar Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM). Sampel yang digunakan dalam penelitian ini adalah isolat Lactobacillus acidophilus yang diperoleh dari Laboratorium Mikrobiologi, Fakultas Kedokteran Universitas Brawijaya, Malang. Konsentrasi ekstak yang digunakan dalam penelitian, yaitu 0%, 50%, 55%, 60%, 65%, 70% dan 75% v/v dengan tiga kali pengulangan. Konsentrasi Lactobacillus acidophilus sebesar 106CFU/ml. Hasil uji statistik menunjukkan bahwa kelompok ekstrak daun pandan memiliki perbedaan yang signifikan (One Way Anova, p>0.05). Uji korelasi Pearson menunjukkan hubungan kuat dengan arah negatif yang dapat diartikan semakin meningkatnya konsentrasi ekstrak maka pertumbuhan bakteri semakin berkurang. Uji regresi menunjukkan efektivitas antibakteri ekstrak daun pandan sebesar 93,4% (R square 0,934). Berdasarkan penelitian ini, dapat disimpulkan bahwa ekstrak daun pandan efektif sebagai antibakteri terhadap Lactobacillus acidophilus secara in vitro.

English Abstract

Dental caries is a demineralization process attacking dental hard tissue caused by microbial activities leaven the carbohydrates. Lactobacillus acidophilus is one of the bacterium that plays an important role in dental caries. Pandan leaves peels are natural substances that can perform as an antibacterial because its suspected to contain active substances such as flavonoid, alkaloid, tannin, and saponin. This research aims to prove the effect of pandan leaves antibacterial toward Lactobacillus acidophilus using in vitro. The experiment of laboratoric experimental is used tube dilution test to determine the Minimum Inhibitory Concentration (MIC) and to determine the Minimum Bacterycidal Concentration (MBC). The sample used in this experiment was Lactobacillus acidophlus obtained from the laboratory of Microbiology, Faculty of Medicine, Brawijaya University, Malang. The extract concentration used were 0%, 50%, 55%, 60%, 65%, 70% and 75% v/v with three repetition. The concentration of Lactobacillus acidophilus was 106CFU/ml. The result of statistic test shows each group of pandan leaves extract carries significant difference (One Way Anova, p>0.05). Pearson correlation test showed a strong correlation with the negative direction that can be interpreted the increasing concentration of the extract, the bacteria growth on the wane. Regression test showed pandan leaves extract antibacterial effectiveness as 93,4% (R square=0,934). Based on this study, it can be concluded that pandan leaves extract is effective as antibacterial toward Lactobacillus acidophilus with in vitro procedure.
