Abstract

Wabah penyakit terus menyebar secara tidak terkontrol mengakibatkan tingginya tingkat kematian ikan sehingga hasil produksi yang diperoleh semakin turun. Serangan virus KHV (Koi Herpes Virus) pada ikan koi (Cyprinus carpio L.) memperoleh persentase kematian sebesar 80-85% dari jumlah biomassa dengan masa inkubasi antara 1-7 hari. Infeksi KHV terjadi pada kulit dan insang juga organ dalam seperti ginjal, hati dan otak karena memiliki prevalensi tinggi dibandingkan dengan organ lain. Tujuan dari penelitian ini adalah untuk mengetahui seberapa besar dampak dari infeksi KHV dan gambaran histopatologi ginjal ikan koi (C. carpio L.) serta mengetahui gejala klinis ikan koi yang terinfeksi KHV. Metode pengambilan data yang digunakan dalam penelitian ini adalah deskriptif kualitatif dengan parameter utama yaitu uji Polymerase Chain Reaction (PCR) pada organ insang dan ginjal ikan koi (C. carpio L.) dan uji histopatologi ginjal ikan koi (C. carpio L.). Parameter penunjang yang digunakan yaitu pengukuran kualitas air meliputi suhu, pH dan DO. Hasil uji PCR menunjukkan ikan koi (C. carpio L.) terinfeksi KHV dan hasil uji histopatologi terdapat beberapa kerusakan pada ginjal ikan antara lain inflamasi, degenerasi hidropik, hemorhagi, dan nekrosis. Gejala klinis ikan terinfeksi KHV seperti berenang dekat permukaan, sistem keseimbangan tubuh hilang dan filament insang pucat. Gejala lain yang dicirikan oleh ikan yaitu produksi lendir berlebih, pendarahan ditutup insang, ekor dan sirip, bercak putih pada kulit, dan enophtalmos atau mata tenggelam. Hasil pengukuran kualitas air suhu sebesar 27,50C, pH 7 (normal) dan DO 4,85 ppm.

English Abstract

The plague of the disease continued to spread uncontrollably resulting in high levels of fish mortality so that the yield was decreasing. The attacks of KHV (Koi Herpes Virus) on koi fish (Cyprinus carpio L.) have a death rate by 80-85% of the amount of biomass with an incubation period of 1-7 days. The KHV infection occurs in fish skin and gills as well as in internal organs such as kidneys, liver, and brain due to the high prevalence compared to other organs. The purpose of this study was to find the impact of KHV infection and the histopathology picture of koi kidneys (C. carpio L.) as well as to know the clinical symptoms of the infected koi fish. The data collection method in this study was a descriptive qualitative with main parameters of Polymerase Chain Reaction (PCR) test on fish kidneys and gills (C. carpio L.) and histopathology test on the kidneys. Moreover, the supporting parameters used were water quality measurements including temperature, pH, and dissolved oxygen. The PCR test results showed that the koi fish (C. carpio L.) was infected with KHV while the results of histopathology test pointed out some damages on the fish kidneys such as inflammation, hydropic degeneration, hemorrhagic, and necrosis. The clinical symptoms of the infected fish were swimming near the surface, loss of body balance system, and pale gill filaments. There were other symptoms characterized by the fish such as excess mucus production, bleeding on gill lid, tail, and fins, white patches on the skin, and enophthalmos or sunken eyes. The results of water quality measurement showed a temperature of 27,50C with pH 7 (normal) and dissolved oxygen of 4,85 ppm.
