Abstract

Burkholderia glumae adalah patogen yang menyebabkan penyakit Bacterial Grain Rot (BGR). Bakteri B. glumae merupakan jenis bakteri yang berstatus Organisme Pengganggu Tumbuhan Karantina (OPTK) A2, artinya penyebarannya masih terbatas di Indonesia. Selain itu, karakterisasi patogen B. glumae juga masih terbatas. Penyakit BGR dapat menyebabkan kerugian hasil hingga 70-75%. Penyebaran patogen B. glumae melalui benih, pencegahan dapat dilakukan dengan perlakuan benih. Perlakuan benih dapat menggunakan bahan kimia sintetik, kimia nabati, dan perlakuan fisik melalui Dry Heat Treatment (DHT). Penelitian ini bertujuan untuk mengetahui karakterisasi bakteri B. glumae, dan mengevaluasi efektifitas perlakuan benih metode kimia, fisik, dan kombinasinya dalam mengendalikan patogen B. glumae yang mengkontaminasi benih padi serta analisis terhadap daya perkecambahan benih padi tersebut. Penelitian dilaksanakan mulai bulan Januari sampai Juli 2017 bertempat di Laboratorium Proteksi Tanaman Departemen Biotechnology PT. BISI International Tbk., Plosoklaten, Kediri, Jawa Timur. Penelitian dilaksanaan dengan beberapa tahapan, pertama adalah pengambilan sampel benih padi yang bergejala B. glumae, isolasi, pemurnian bakteri target kemudian identifikasi berdasarkan karakter morfologi, biokimia dan teknik PCR. Benih padi yang diduga terinfeksi bakteri B. glumae akan digunakan untuk tahap penelitian kedua, yaitu uji efektifitas perlakuan benih dengan minyak cengkeh, minyak serai wangi, HCl, TSP, Fungisida berbahan aktif Tembaga hidroksida, DHT dan kombinasinya dalam mengendalikan patogen B. glumae pada benih padi serta pengaruh terhadap mutu fisiologis benih tanaman padi. Hasil penelitian menunjukkan bahwa perlakuan benih tunggal dengan DHT 56 oC selama 15 menit, minyak serai wangi 1% perendaman 30 menit, TSP 2 gram/ 10 ml perendaman 5 jam, Fungisida berbahan aktif Tembaga hidroksida 2 gram/ liter perendaman 5 jam dan perlakuan kombinasi TSP 2 gram/ 10 ml perendaman 5 jam + HCl 0,25 N perendaman 15 menit, DHT 56 oC selama 15 menit + Fungisida berbahan aktif Tembaga hidroksida 2 gram/ liter perendaman 5 jam efektif untuk menghambat perkembangan bakteri B. glumae dan tidak menurunkan mutu fisiologis benih.

English Abstract

Burkholderia glumae is a pathogen the it cause disease of Bacterial Grain Rot (BGR). Bacteria B. glumae is a type of bacteria that is a Plant Quarantine Pest Organism (OPTK) A2, meaning the species is limited in Indonesia. In addition, the characterization of the pathogen B. glumae also limited. The disease can cause harm BGR results up to 70-75%. The spread of pathogens B. glumae through seeds, prevention can be done by seed treatment. Seed treatment can use synthetic chemicals, chemical plant, and physical treatment through Dry Heat Treatment (DHT). The purpuse of this study is to know characterization of bacteria B. glumae, and evaluating the effectiveness of seed treatment method of chemical, physical, and combination in controlling pathogens B. glumae that contaminate rice seeds as well as analysis of the rice seed germination power. This study was conducfed at Department of Plant Protection Laboratories Biotechnology PT BISI International Tbk., Plosoklaten, Kediri, East Java from January to July 2017. The stages performed in this study were sampling rice seed syptoms B. glumae, isolation, purification of the bacteria then identification is based on the character of the morphology, biochemistry and PCR techniques. The rice seeds infected with bacteria allegedly B. glumae to be used for a second research phase, i.e., test the effectiveness of seed treatment with clove oil, Lemongrass oil fragrant, HCl, TSP, Fungicides made from Active Copper hydroxide, DHT and combination in controlling pathogens B. glumae on rice seeds and the effect onthe physiological quality of seeds of rice plant. The results showed that a single seed treatment with DHT 56 oC for 15 minutes, fragrant Lemongrass oil 1% soaking 30 minutes, TSP 2 grams/ 10 ml 5 hours of soaking, Fungicides made from Active Copper hydroxide 2 grams/ liter 5 hours of soaking and treatment combination of TSP 2 grams/ 10 ml soaking 5 hours + HCl 0.25 N 15 minutes of soaking, DHT 56 oC for 15 minutes + Active Copper hydroxide-based Fungicide 2 grams/liter of 5 hours of soaking to effectively inhibit bacterial development B. glumae and does not degrade the quality of physiological seed.
