Abstract

Produksi budidaya udang vaname di Indonesia tahun 2010 meningkat 12,69% sedangkan permintaan eskpor menurun menjadi 2,77%. Penurunan permintaan ekspor disebabkan karena bakteri luminescence Pseudomonas fluorescens. Tinta cumi-cumi merupakan salah satu bahan alami yang dapat digunakan sebagai antibakteri karena adanya senyawa fenol. Metode penelitian yang digunakan adalah metode eksperimen dengan Rancangan Acak Lengkap (RAL) menggunakan 5 perlakuan dan 3 kali ulangan. Dosis ekstrak tinta cumi-cumi yang digunakan yaitu A (1,25 ppm), B (2,5 ppm), C (3,75 ppm), D (5 ppm). Parameter utama dalam penelitian ini yaitu biofilm coverage rate (BCR) dan visual 3 dimensi biofilm bakteri P. fluorescens pada karapas post larva udang vaname. Parameter penunjang yaitu pengukuran kualitas air meliputi suhu, pH, DO, salinitas, nitrit dan nitrat. Berdasarkan hasil penelitian didapatkan rerata nilai BCR yang terbaik pada perlakuan C yaitu 7,32% dinyatakan berpengaruh sangat nyata terhadap nilai BCR pada karapas post larva udang vaname. Hubungan dosis ekstrak tinta cumi-cumi terhadap nilai BCR bakteri P. fluorescens pada karapas post larva udang vaname mendapatkan persamaan linier yaitu y = 20,985 – 2,7131x dengan R2 = 0,92. Hasil visual 3 dimensi menunjukkan bahwa perlakuan C memiliki tutupan biofilm terendah. Hasil parameter kualitas air yaitu suhu 24–26 0C, pH 7,50–8,15, DO 4,51–5,63 mg/L, salinitas 24–26 ppt, nitrit 0–5 mg/L dan nitrat 0–25 mg/L. Kesimpulan dari penelitian ini yaitu ekstrak tinta cumi – cumi berpengaruh terhadap nilai BCR dan visual 3 dimensi biofilm bakteri P. fluorescens pada karapas post larva udang vaname dengan perlakuan terbaik adalah perlakuan C (3,75 ppm).

English Abstract

Production of vaname shrimp cultivation in Indonesia in 2010 increased by 12.69% while export demand decreased to 2.77%. The decrease in export demand is due to the bacteria luminescence Pseudomonas fluorescens. Squid ink is one of the natural ingredients that can be used as antibacterial because of the compound of phenol. The method used in this research was experimental method with Completely Randomized Design (RAL) using 5 treatments and 3 replications. The dosage of squid ink extract were A (1.25 ppm), B (2.5 ppm), C (3.75 ppm), D (5 ppm). The main parameters in this study are biofilm coverage rate (BCR) and visual 3-dimensional biofilm of P. fluorescens on post larva carapace of vaname shrimp. The supporting parameters were water quality measurement included temperature, pH, DO, salinity, nitrite and nitrate. The results showed that the best average value of BCR at the treatment of C was 7,32% that squid ink extract had a significant effect on BCR value on post larva carapace of vaname shrimp. The relation of dose of squid ink extract to BCR value of P. fluorescens bacteria on post larva carapace of vaname shrimp obtained linear equation y = 20,985 - 2,7131x with R2 = 0,92. 3-dimensional visual results showed that treatment C has the lowest biofilm cover. The results of water quality parameters for temperature was 24-26 0C, pH 7,50-8,15, DO 4,51-5,63 mg/L, salinity 24-26 ppt, nitrite 0-5 mg/L and nitrate 0-25 mg/L. The conclusion of this research is squid ink extract influenced to BCR value and visual 3 dimension of biofilm of P. fluorescens bacteria on post larva carapace of vaname shrimp with the best treatment was on C (3,75 ppm) treatment.
