Abstract

Rhodamin B dan sakarin merupakan zat xenobiotik yang berbahaya bagi tubuh. Konsumsi rhodamin B dan sakarin secara terus menerus dapat menyebabkan kanker serta gangguan beberapa fungsi organ seperti hati, saluran pernafasan, dan saluran pencernaan. Penelitian ini dilakukan untuk mengetahui pengaruh toksisitas rhodamin B dan sakarin terhadap kadar MDA dan gambaran histopatologi lambung tikus putih (Rattus norvegicus). Tikus putih (Rattus norvegicus) dibagi menjadi 4 kelompok perlakuan, yaitu kelompok K(-), P1, P2, dan P3. Kelompok K(-) tidak diberikan rhodamin B dan sakarin, kelompok P1 diberikan rhodamin B dengan dosis 22,5 mg/kgBB, kelompok P2 diberikan sakarin dengan dosis 157,77 mg/kgBB, dan kelompok P3 diberikan kombinasi rhodamin B dengan dosis 22,5 mg/kg BB ditambah sakarin dengan dosis 157,77 mg/kgBB. Pemberian rhodamin B dan sakarin dilakukan selama 30 hari per-oral menggunakan sonde lambung. Parameter yang diamati adalah kadar MDA dengan menggunakan uji Asam Tiobarbiturat (TBA) dan gambaran histopatologi lambung tikus putih (Rattus norvegicus) dengan menggunakan pewarnaaan Hematoksilin Eosin (HE). Analisa kadar MDA dilakukan secara kuantitatif menggunakan analisa ragam ANOVA dilanjutkan dengan uji Tukey (Beda Nyata Jujur) dengan α = 0,05 dan data gambaran histopatologi lambung dilakukan secara deskriptif kualitatif. Hasil penelitian menunjukkan pengaruh toksisitas rhodamin B dan sakarin terhadap kadar MDA dan gambaran histopatologi lambung. Pada kelompok P3 memberikan pengaruh lebih toksik ditandai dengan kadar MDA tertinggi dibanding kelompok lain, yaitu 2,799 ± 0,309. Gambaran histopatologi lambung pada kelompok P3 menunjukkan adanya erosi, hemoragi, infiltrasi sel radang, karioreksis, kariolisis, dan piknosis.

English Abstract

Rhodamine B and saccharin are xenobiotic substances that are harmful to the body. The continuous consumption of rhodamine B and saccharin can lead to cancer and disruption of some organ functions such as the liver, respiratory tract, and gastrointestinal tract. This study was conducted to determine the toxicity effect of rhodamine B and saccharine on gastric MDA levels and histopathology in white rat (Rattus norvegicus). The white rat (Rattus norvegicus) were divided into 4 treatment groups: K(-), P1, P2, and P3 group. K(-) group were not given rhodamine B and saccharin, P1 group were given rhodamine B at dose of 22,5 mg/kgBW, P2 group were given saccharin at dose of 157,77 mg/kgBW, and P3 group were given the combination of both of it. The administration of rhodamine B and saccharin was performed for 30 days per-orally. The parameters which observed in this reseach were gastric’s MDA level with Tiobarbituric Acid (TBA) test and gastric histopathology of white rat (Rattus norvegicus) with Hematoxylin Eosin (HE) stained. Measurement of gastric MDA level was analyzed statistically quantitative conducted by ANOVA and followed by Fisher’s exact test with α = 0,05. Gastric histopathology was analyzed in descriptive qualitativeThe result showed that there were differences of rhodamine B and saccharin administration effects to MDA levels and gastric histopathology. P3 group showed the most toxic effect marked by highest MDA levels compared to the other groups of 2.799±0,309. Other than that erotion, hemorrhage, inflammatory cell infiltration, karyorrhexis, karyolysis, and pyknosis were found in gastric histopathology in P3 group.
