Abstract

Minyak atsiri dari tanaman adas pahit (Foeniculum vulgare Mill. var. vulgare) merupakan salah satu minyak atsiri yang mempunyai potensi produksi. Minyak atsiri dapat diisolasi menggunakan metode distilasi uap. Salah satu faktor yang mempengaruhi proses distilasi adalah waktu distilasi. Waktu distilasi mempengaruhi kualitas dan profil komponen minyak atsiri. Profil komponen mempengaruhi aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui pengaruh waktu distilasi terhadap rendemen, sifat fisik, profil komponen minyak atsiri biji adas pahit dan aktivitasnya sebagai antibakteri Staphylococcus aureus. Isolasi minyak atsiri biji adas pahit dilakukan menggunakan metode distilasi uap 5, 7, dan 9 jam. Karakterisasi sifat fisik minyak atsiri biji adas pahit dilakukan berdasarkan penentuan wujud, warna, aroma, indeks bias, dan berat jenis. Analisis minyak atsiri biji adas pahit dilakukan menggunakan Kromatografi Gas-Spektrometri Massa (KG-SM). Uji aktivitas antibakteri S. aureus dari minyak atsiri biji adas pahit menggunakan metode difusi cakram. Minyak atsiri biji adas pahit yang diperoleh berupa cairan berwarna kuning dengan rendemen tertinggi (0,72%) diperoleh dari hasil distilasi 9 jam. Analisis KG-SM menunjukkan minyak atsiri biji adas pahit dengan waktu distilasi 5, 7, dan 9 jam menghasilkan berturut-turut 10, 4, dan 4 senyawa. Empat komponen utama minyak atsiri biji adas pahit adalah estragol, fenkon, α-pinen, dan limonen. Kadar estragol tertinggi (87,93%) diperoleh dari hasil distilasi uap 7 jam. Uji aktivitas antibakteri S. aureus dari minyak atsiri biji adas pahit menunjukkan diameter zona hambat sebesar 11-12 mm, tergolong kategori resisten. Waktu distilasi mempengaruhi rendemen, profil komponen dan aktivitas antibakteri S. aureus dari minyak atsiri biji adas pahit. Namun tidak mempengaruhi sifat fisik minyak atsiri biji adas pahit.

English Abstract

Essential oil from bitter fennel (Foeniculum vulgare Mill. var. vulgare) is one of the essential oils that has potential production. Essential oils can be isolated by steam distillation. One of the factors affecting distillation is distillation time. Distillation time affects the quality and component profile of essential oils. The component profile affects antibacterial activity. The aims of this research are to know the influence of distillation time on yield, physical properties, component profile of bitter fennel seeds oils and its antibacterial activity against Staphylococcus aureus. Bitter fennel seeds oils were isolated by steam distillation for 5, 7, and 9 hours. The physical properties of its essential oils were determined based on physical state and appearance, color, odor, refractive index, and specific gravity. The chemical compounds were analyzed by Gas Chromatography-Mass Spectrometer (GC-MS). The antibacterial activity of bitter fennel seeds oils against S. aureus were performed by disc diffusion. The result in this research indicates that bitter fennel seeds oils is a yellow liquid. The highest yield of the essential oils (0.72%) is obtained by distillation for 9 hours. The result of GC-MS showed that bitter fennel seeds oils isolated by steam distillation for 5, 7, and 9 hours contained 10, 4, and 4 compounds, respectively. Four main components of fennel seeds oils are estragole, fenchone, α-pinene, dan limonene. The highest content of estragole (87.93%) is obtained by steam distillation for 7 hours. The antibacterial test of bitter fennel seeds oils against S. aureus shows the diameter of zone inhibition at 11-12 mm, classified as resistant category. The distillation time influences the yield, component profile and antibacterial activity of bitter fennel seeds oils against S. aureus, but it does not influence the physical properties of bitter fennel seeds oils.
