Abstract

Tanaman kubis (Brassica oleracea var. Capitata L.) merupakan salah satu tanaman holtikultura familia Brassicacea yang banyak dibudidayakan di Indonesia. Salah satu faktor yang menyebabkan menurunnya produksi kubis di Indonesia adalah penyakit busuk hitam yang disebabkan oleh bakteri Xanthomonas campestris pv. campestris. Berbagai upaya pengendalian telah dilakukan oleh para petani kubis, baik pengendalian secara kimia, fisik dan biologi. Tetapi petani kubis lebih banyak melakukan pengendalian dengan cara kimia yaitu dengan menggunakan pestisida. Kondisi ini memberikan gagasan untuk melakukan pengendalian dengan menggunakan agens hayati yang bersifat ramah lingkungan. Salah satu agens hayati yang dapat dimanfaatkan untuk mengendalikan penyakit pada patogen tanaman yaitu bakteri yang bersifat antagonis. Tujuan dari penelitian ini adalah untuk memperoleh isolat bakteri rizosfer dari lahan kubis PHT dan konvensional yang bersifat antagonis. Hipotesis pada penelitian ini yaitu pada lahan kubis PHT ditemukan lebih banyak jenis bakteri yang bersifat antagonis terhadap penyakit busuk hitam dibandingkan dengan lahan kubis konvensional. Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya Malang mulai bulan Januari 2017 sampai April 2017. Metode penelitian terdiri dari metode survei dan metode percobaan. Penelitian terdiri dari 8 perlakuan dengan masing-masing perlakuan diulang sebanyak 4 kali. Variable pengamatan pada penelitian ini yaitu kelimpahan bakteri rizosfer, keanekaragaman bakteri rizosfer, dan indeks penghambatan bakteri antagonis terhadap bakteri X. campestris pv. campestris. Data pengamatan dianalisis menggunakan analisis ragam pada taraf 5%. Apabila hasil pengujian terdapat perbedaan yang nyata maka dilanjutkan dengan uji Duncan taraf 5%. Identifikasi terhadap keenam isolat bakteri rizosfer yang bersifat antagonis terhadap bakteri X. campestris pv. campestris berdasarkan buku pedoman Bergey’s Manual of Determinative (1994) dan Schaad et al., (2001) menunjukkan bahwa empat isolat bakteri dengan kode P-A1, P-A5, P-B1, dan P-C2 termasuk kedalam genus Pantoea, satu isolat bakteri dengan kode P-A2 masuk kedalam genus Erwinia dengan kemampuan penghambatan tertinggi, dan satu isolat bakteri dengan kode K-D masuk kedalam genus Bacillus.

English Abstract

Cabbage (brassica oleracea var. Capitata L) is one of the holticultural plants of Brassicacea familia that is cultivated a lot in Indonesia. One of the factor that decrease the productivity of cabbage is caused by the black rot disease due to bacteria of Xantomonas campestris pv. Campestris. Some efforts have been done by the cabbage farmers, such as chemically, phisically, and biologically controls. However, the cabbage farmers prefer to use chemical way by using pesticides. This condition brings out the idea to do the biogically control by using biological agents which is eco-friendly. One of the biological agent that can be used to control the disease on the plants pathogens is an antagonist bacteria. The purpose of this research is to get the isolates of rhizosphere bacteria from the land of PHT cabbage and conventional cabbage which is antagonistic. The hypothesis of this research is the PHT cabbage land was found a lot of antagonistic bacteria to againts the black rot disease rather than in conventional cabbage land. The research was conducted in the Laboratory of Plant Pathology, Faculty of Agriculture, University of Brawijaya, Malang started from January 2017 until April 2017. The research method consists of survey method and experimental method. The research consist of 8 treatments which each treatment was repeated 4 times. The varieable of this research are the amount of rhizosphere bacteria, the variety of rhizosphere bacteria, and index of obstacle between antagonist bacteria to X campestris pv. Campestris bacteria. The data was analized by using analysis of variety on 5% level. If this research has a significally different result, so it will be continued by using Duncan test on the 5% level. Identification of six rhizosphere bacterial isolates that are antagonistic to bacteria X. campestris pv. campestris based on Bergey's Manual of Determinative (1994) and Schaad et al. (2001) indicated that four isolates bacteria with P-A1, P-A5, P-B1 and P-C2 isolates code that include to the genus of Pantoea, then one isolates bacteria with P-A2 isolates code include to the genus of Erwinia , as the highest resistor, and one isolates bacteria with K-D isolates code include to the genus of Bacillus sp.
