Pengaruh Pemberian Ekstrak Kasar Daun Kecubung (Datura Metel L.) Terhadap Daya Hambat Bakteri Vibrio Alginolyticus Secara In Vitro

Abstract

Perkembangan budidaya baik udang maupun ikan semakin lama semakin pesat, akan tetapi terdapat beberapa kendala dalam budidaya salah satunya adalah serangan penyakit yang bersifat bakterial yaitu penyakit vibriosis yang disebabkan oleh bakteri V. alginolyticus. Penggunaan antibiotik banyak digunakan untuk mengobati penyakit tersebut, akan tetapi penggunaan secara terus menerus akan mengakibatkan resistensi dan pencemaran lingkungan. Untuk mengatasi permasalahan tersebut dapat menggunakan bahan alami, salah satunya adalah daun kecubung (D. metel L.). Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak kasar daun kecubung (D. metel L.) terhadap daya hambat bakteri V. alginolyticus secara in vitro. Metode yang diguanakan adalah metode eksperimental dengan rancangan percobaan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan yaitu perlakuan A (75 ppm), B (150 ppm), C (225 ppm), D (300 ppm) dan E (375 ppm) yang dilakukan 3 kali pengulangan. . Hasil penelitian menunjukkan bahwa zona hambat tertinggi terdapat pada perlakuan E (375 ppm) dengan didapatkan rata-rata zona hambat sebesar 6,79 mm sedangkan zona hambat terendah terdapat pada perlakuan A (75 ppm) yang didapatkan rata-rata zona hambat dengan ukuran 3,83 mm. Hubungan zona hambat antar perlakuan ekstrak kasar daun kecubung (D. metel L. )terhadap bakteri V. alginolyticus menunjukkan perpotongan garis secara linear dengan persamaan y = 0,0096x + 2,698 dengan koefisien nilai determinasi (R2) sebesar 0,916 dan nilai korelasi sebesar 0,96, dimana derajat keeratan hubungan antara dosis ekstrak daun kecubung (D. metel L.) dan nilai rata-rata zona hambat termasuk dalam kategori erat. Hubungan antara pemberian ekstrak kasar daun kecubung (D. metel L.) dalam menghambat pertumbuhan bakteri V. alginolyticus dengan zona hambat yang dihasilkan menunjukkan respon yang sedang. Ekstrak kasar daun kecubung (D. metel L.) terbukti mampu membunuh bakteri V. alginolyticus.

English Abstract

The development of aquaculture of both shrimps and fish are getting more rapidly, but there are some problems in aquaculture, one of them is a bacterial disease. One of bacterial disease that is caused by the bacterium of V. alginolyticus. The antibiotics are mostly used to treat the disease, but the continuous usage will be inflict of resistance. One way to overcome these problems is using medicinal and natural plants, one of which is kecubung leaves (D. metel L.). This study was aimed to determine the effects of giving between crude extracts of kecubung leaves (D. metel L.) to inhibit of V. alginolyticus, in vitro. This research used experimental method, using 5 treatments which were treatment A (75 ppm), B (150 p pm), C (225 ppm), D (300 ppm) and E (375 ppm), and 3 repetition . The results showed that the highest inhibitory zone was on treatment E (375 ppm) with inhibition zone of 6,79 mm. Meanwhile, the lowest treatment A (75 ppm) with inhibition zone of 3,83 mm. Relations of inhibition zones between crude extracts of kecubung leaves (D. metel L.) treatments to V. alginolyticus showed liniarly crossover y = 0,0096x + 2,698 with coefficient value of determination (R2) of 0,916 and coefficient value of colleration of 0,96. The relation between crude extracts of kecubung leaves (D. metel L.) and inhibiting the growth of V. alginolyticus resulted a moderate response. Crude extracts of kecubung leaves (D. metel L.) had proved to kill V. alginolyticus.
