Abstract

Aterosklerosis merupakan penyakit yang disebabkan karena terbentuknya plak di dinding arteri. Penyakit aterosklerosis dapat menyebabkan iskemia dan infark jantung, stroke, hipertensi renovaskuler, dan penyakit oklusi tungkai bawah. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian diet aterogenik terhadap kadar kolesterol dan gambaran histopatologi aorta pada kelinci New Zealand White. Diet aterogenik diberikan sebanyak 40 g/ekor/hari selama 28 hari. Penelitian ini menggunakan metode pre dan post test examination. Kelinci New Zealand White dibagi menjadi 2 kelompok yaitu kontrol negatif (A) diberi diet normal dan kontrol positif (B) diberi diet aterogenik. Setiap kelompok terdiri dari 8 ekor kelinci. Kadar kolesterol serum diukur dengan menggunakan metode spektrofotometri serta gambaran histopatologi aorta dengan menggunakan pewarnaan Hematoxylin Eosin (HE). Analisa statistik menggunakan SPSS rev.20,0 dengan uji t tidak berpasangan (Independent t test) untuk data kadar kolesterol sedangkan pengamatan histopatologi aorta dengan analisa perbandingan dengan jaringan normal. Hasil penelitian menunjukkan pemberian diet aterogenik selama 28 hari mampu meningkatkan kadar kolesterol serum sebesar 513,5% secara signifikan (p<0,05) dan pada gambaran histopatologi aorta tampak perubahan struktur sel endotel disertai dengan awal pembentukan foam cell.

English Abstract

Atherosclerosis is a condition caused by formation of plaque in the arterial wall. Atherosclerosis can caused ischemia, heart infarct, stroke, renovascular hypertension, and the occlusion of lower extremity. The purpose of this study was to determined the effect of atherogenic diet induced to the cholesterol level and histopathology of aorta in New Zealand White rabbit. Atherogenic diet given 40 g/rabbit/day orally for 28 days. The study used pre and post examination test methods. Rabbits were divided into 2 groups; negative control (A) induced by normal diet and positive control (B) induced by atherogenic diet. Each groups consists of 8 rabbits. Cholesterol level serum was measured by spectophotometric method and the histopathology of aorta was observed by Hematoxylin Eosin (HE) staining. Statistic analysis used SPSS rev.20.0 with independent t-test for the cholesterol level and qualitative observation method for the histopathology of aorta. The results of this study showed the atherogenic diet induced for 28 days significantly (p<0,05) increase the cholesterol level for 513,5 % and the histopathology of aorta showed the changes in endothelial cells with the early formation of foam cell.
