Abstract

Kanker mammae merupakan tumor ganas yang tumbuh di dalam jaringan mammae yang dapat dipicu oleh agen karsinogenik DMBA (Dymethylbenz(α) Anthrasen). Kanker mammae akibat induksi DMBA mempunyai kemungkinan menyebar ke uterus. Pengobatan kanker selain dengan operasi dan kemoterapi juga bisa menggunakan herbal. Kurkumin merupakan senyawa polifenik yang mempunyai efek sebagai antiproliferasi, pemacu apoptosis dan antioksidan. Vitamin E merupakan antioksidan primer yang mampu memutus reaksi pembentukan radikal bebas. Tujuan penelitian ini adalah untuk mengetahui pengaruh terapi kombinasi Kurkumin dengan Vitamin E pada ekspresi iNOS dan BCL-2 pada uterus tikus model kanker mammae yang diinduksi DMBA. Pembuatan hewan model kanker mammae dilakukan dengan induksi DMBA dosis 10 mg/kg BB dan estrogen dosis 20.000 IU/kg BB. Penelitian ini dibagi 5 kelompok perlakuan yaitu kelompok kontrol negatif (KN), kontrol positif (KP), perlakuan satu (P1) terapi dengan Kurkumin 48mg/kg BB dan Vitamin E 300 IU/ekor, perlakuan dua (P2) dosis 72mg/kg BB dan 200 IU/ekor, dan perlakuan tiga (P3) dosis 108mg/kg BB dan 100 IU/ekor. Ekspresi iNOS dan BCL-2 diamati dengan Imunohistokimia dan dianalisis dengan uji ANOVA dan dilanjutkan dengan uji Tukey (α = 0,05). Hasil penelitian menunjukkan bahwa terapi kombinasi Kurkumin dan Vitamin E berpengaruh nyata terhadap penurunan ekspresi iNOS dan BCL-2 pada uterus tikus model kanker mammae.. Dosis efektif yaitu 72 mg/kg BB kurkumin dan 200 IU/ekor vitamin E yang mampu menurunkan ekspresi iNOS sebesar 20,29% dan menurunkan ekspresi BCL-2 sebesar 61,65%. Pemberian kombinasi Kurkumin dan Vitamin E dapat digunakan sebagai terapi kanker mammae yang memberikan efek pada uterus.

English Abstract

Mammary cancer is malignant tumor that grow inside mammary tissue and can be triggered by carcinogenic agent DMBA. Mammary cancer induced by DMBA have possibility to spread to uterus. Another way to stop cancer besides surgery and chemotherapy can be supported by herbal therapy. Curcumin is one of the polyphenols type that has an effect as antiproliferation, trigger apoptosis and antioxidant. Vitamin E is a primary antioxidant that can cut reaction of free radical formation. The purpose of this research was to discover the effect of combination of curcumin with vitamin E toward expression of INOS and BCL-2 in uterus of rats mammary cancer model induced by DMBA. The development of rats mammary cancer model was done by induction of DMBA with dose 10mg/kg bodyweight and estrogen with dose 20.000 IU/kg bodyweight. This research divided 5 groups that is Negative Control Group, Positive Control Group, First Treatment (P1) used 48 mg/kg bodyweight curcumin and vitamin E 300 IU each rat, second Treatment (P2) used 72 mg/kg bodyweight curcumin and vitamin E 200 IU each rat, third Treatment (P3) used 108 mg/kg bodyweight curcumin and vitamin E 100 IU each rat. The expression of INOS and BCL-2 were monitored using immunohistochemistry method. Data analysis was done by one way ANOVA test and continued by Tukey test (α = 0,05). The result of the research showed that combination of curcumin and vitamin E significantly reduced expression of INOS and BCL-2 in uterus of rat mammary cancer. The most effective dose is curcumin with 72 mg/kg of bodyweight and vitamin E with dose 200 IU each rat that have an ability to decrease 20.29% of iNOS expression and decrease 61.65% of BCL-2 expression. As conclusion, combination of curcumin and vitamin E can become one of mammary cancer therapy that can affect to uterus.
