Abstract
Kaptopril sering digunakan pada terapi hipertensi. Kaptopril memiliki kelarutan yang tinggi dengan permeabilitas rendah menyebabkan bioavailabilitasnya rendah. Penelitian ini bertujuan untuk menguji pengaruh konsentrasi guar gum terhadap profil pelepasan kaptopril dari kapsul FDDS effervescent dengan metode pembuatan granulasi basah. Konsentrasi guar gum yang digunakan yaitu 7,5%(FA), 15%(FB) dan 22,5%(FC). Sudut diam granul ketiga formula berkisar antara 22,22-27,28° yang menunjukkan ketiganya memiliki sifat alir yang baik. Total waktu terapung kapsul ketiga kurang dari 8 jam, dimana FA terapung selama 5 jam sedangkan FB dan FC terapung selama 7 jam. Perbedaan persentase pelepasan dianalisis dengan repeated ANOVA. Hasil analisis menunjukkan ketiga formula memiliki perbedaan pelepasan kaptopril yang signifikan pada jam ke-6 (p<0,05) dengan persen pelepasan FA, FB, dan FC sebesar 106,616±3,564%, 101,823±2,858%, dan 94,148±0,746%. Sedangkan pelepasan jam ke-7 menunjukkan pelepasan FB terhadap FC tidak signifikan (p>0,05). Hasil analisis statistik efisiensi disolusi (ED) 5 jam menunjukkan signifikansi FC terhadap FA dan FB (p<0,05). Namun ED5 FB tidak signifikan terhadap FA (p>0,05) dengan FA memiliki ED5 paling tinggi. Hasil analisis kinetika pelepasan obat menunjukkan ketiga formula mengikuti kinetika orde nol dengan mekanisme transpor super case II. Dari penelitian ini dapat disimpulkan peningkatan konsentrasi guar gum dapat memperlama pelepasan kaptopril.
English Abstract
Captopril is widely used in hypertension therapy. Captopril has highly soluble with low permeability caused low bioavailability. This study aims to analyze the effect of concentration captopril on release profile of FDDS effervescent capsule of captopril by wet granulation method. Three different concentration guar gum which used were 7,5% (FA), 15% (FB), and 22,5% (FC) . The values of angle of repose were found in between 22,22-27,28° that showed all three formulas had good flowability. From the bouyancy studies, it showed that capsule FA, FB, and FC floated less than 8 hours, which FA floated in 5 hours, FB and FC floated in 7 hours. The different captopril rekease percentage of three formulas analyzed by repeated ANOVA. It showed significant release of captopril (p<0,05) in 6th hours with release percentage FA, FB, and FC were 106,616±3,564%, 101,823±2,858%, dan 94,148±0,746%. The release profile of captopril in 7th hours showed release profile of FB and FC is not significant (p>0,05). Dissolution Efficiency (DE) in 5 h showed FC significant than FA and FB (p<0,05) but FB not significant than FA (p>0,05). The highest DE5 is FA. The result of release profiles inserted in various kinetic equation to analyze the release pattern of the drug and we found that drug release followed zero-order’s kinetics with super case II transport mechanism. Based on the results of this study, it can be concluded that increasing of guar gum concentration can prolonge the release profile of captopril.
