Abstract

Rizosfer telah diketahui banyak mengandung mikroba menguntungkan, salah satunya berasal dari golongan bakteri yaitu Plant Growth Promoting Rhizobacteria (PGPR). Contoh lingkungan yang diduga terdapat PGPR adalah Taman Nasional Bromo Tengger Semeru (TNBTS). Salah satu kawasan di TNBTS adalah Laut Pasir Berbisik. Dikarenakan cekaman lingkungan, tidak banyak ditemukan vegetasi namun pada beberapa titik terdapat tumbuhan alang-alang (Imperata cylindrica (L.) Beauv) yang mampu tumbuh. Diduga terdapat mikroba unggul dari golongan bakteri yang menunjang pertumbuhan alang-alang tersebut. Pada prinsipnya, PGPR dapat dimanfaatkan pada seluruh komoditas pertanian, contohnya padi (Oryza sativa L.). Padi seringkali menghadapi tantangan serangan patogen. Xanthomonas oryzae pv. oryzae (Xoo) dilaporkan sebagai penyebab penyakit hawar daun yang mampu menurunkan produksi mencapai 35,6%. Tujuan penelitian ini adalah memperoleh isolat PGPR dari rizosfer alang-alang di TNBTS serta mengevaluasi potensinya sebagai agens antagonis terhadap Xoo. Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan 1, Fakultas Pertanian, Universitas Brawijaya pada bulan Maret sampai Mei 2017. Penelitian terdiri dari beberapa tahapan yaitu pengambilan sampel dan isolasi bakteri rizosfer, seleksi bakteri rizosfer sebagai PGPR dan agens antagonis, uji antagonis terhadap Xoo secara in vitro, dan karakterisasi serta identifikasi bakteri bakteri hasil seleksi. Hasil eksplorasi menunjukkan bahwa terdapat 46 isolat bakteri berasosiasi dengan rizosfer alang-alang di kawasan TNBTS, 34 isolat diantaranya berpotensi menambat nitrogen, 21 isolat berpotensi melarutkan fosfat, dan 16 isolat mampu melakukan keduanya. Dari 16 isolat bakteri tersebut, 11 isolat berpotensi antagonis terhadap Xoo. Hasil identifikasi secara fisiologi dan biokimia terhadap lima isolat bakteri hasil seleksi menunjukkan bahwa isolat kode 5, 8, dan 12 tergolong genus Pantoea sp. dan isolat 6 dan 10 tergolong genus Erwinia sp. Kelima isolat bakteri memiliki potensi antagonis terhadap Xoo dengan kategori rerata diameter penghambatan sangat kuat (6), kuat (10, 12, dan 8), dan sedang (5).

English Abstract

Rizosphere has been known to contain many beneficial microbes, one of them comes from the bacteria class which is Plant Growth Promoting Rhizobacteria (PGPR). The example of suspected environment which contain PGPR is Bromo Tengger Semeru National Park (BTSNP). One of the area in BTSNP is the sandsea. Due to the extreme environment, there is not many vegetation found there. However, there is one vegetation that is able to grow namely grass (Imperata cylindrica (L.) Beauv). Presumably, there are superior microbes from the class of bacteria that support the growth of the grass. In principle, PGPR can be utilized on all agricultural commodities, for example is rice (Oryza sativa L.). Xanthomonas oryzae pv. oryzae (Xoo) was reported as a cause of leaf blight disease that can reduce production up to 35.6%. The purpose of this study is to obtain PGPR isolates from the rhizosphere of grass in TNBTS and to evaluate its potency as an antagonistic agent against Xoo. The research was conducted at Plant Disease Laboratory 1, Faculty of Agriculture, University of Brawijaya from March to May 2017. The research consisted of several stages: sampling and isolation of rhizospheric bacteria, selection of rhizospheric bacteria as PGPR and antagonist agent againts Xoo, in vitro antagonistic test of bacteria againts Xoo, and characterization and identification of selected bacteria. The results of the exploration showed that there were 46 bacterias associated with rhizosphere of the grass in BTSNP area, 34 isolates had the potential to inhibit nitrogen and 21 isolates had the potential to dissolve phosphate. From 16 selected bacterial isolates that can both inhibit nitrogen and dissolve phospate, 11 isolates potentially had antagonistic activity againts Xoo. The results of physiological and biochemical identification of five selected bacterial isolates showed that bacteria code 5, 8, and 12 belong to the genus Pantoea sp. whereas bacteria code 6 and 10 belong to the genus Erwinia sp. The five bacterial isolates had antagonists activity against Xoo with very strong (6), strong (10, 12, and 8), and average (5) category.
